Descripción general

Thrombin enzyme (Factor IIa, EC 3.4.21.5), a Serine protease, is an important clotting factor in the coagulation cascade that involves the conversion of soluble fibrinogen to insoluble active fibrin strands. In this pathway, Prothrombin is proteolytically converted into an active Thrombin.[1][2]

Idoneidad

Suitable for determining the activity of pure Thrombin and for detecting the activity of Thrombin in plasma.

Principio

Thrombin activity fluorometric assay kit utilizes the ability of thrombin to proteolytically cleave a synthetic substrate and release a fluorophore, 7-amino-4-methylcoumarin (AMC), which can be easily quantified by fluorescence. This assay kit is simple, rapid and can detect thrombin activity as low as 1ng in samples.

Preguntas

  1. Hey just wondering how should we detect the AMC fluorophore released by the substrate? We have flow cytometer in the lab, but not sure how we can detect it.

    1. Please review the protocol at the link below:
      https://www.sigmaaldrich.com/deepweb/assets/sigmaaldrich/product/documents/176/154/mak242bul.pdf

This kit is not intended for use in flow cytometry. A typical measurement is done on a plate reader like those presented in the link below:
