Descripción general

¡Ya está disponible el nuevo kit de ensayo AST, MAK467! La aspartato aminotransferasa (AST), antes denominada glutamato-oxaloacetato transaminasa sérica (SGOT), es una enzima dependiente del fosfato de piridoxal (PLP) que cataliza la conversión de aspartato y α-cetoglutarato en oxaloacetato y glutamato. Igual que ocurre con la alanina aminotransferasa (ALT), los niveles de AST en sangre se utilizan normalmente como marcador de la función hepática. Sin embargo, la AST tiene una distribución tisular más amplia que la ALT y las perturbaciones en los valores de AST pueden ocurrir en respuesta a enfermedades o lesiones en múltiples tejidos, incluido el esquelético y el corazón.

El kit de análisis de actividad de la AST se ha utilizado para determinar la actividad de la aspartato aminotransferasa en muestras.[1][2]

Principio

El kit de análisis de actividad de la aspartato aminotransferasa proporciona un procedimiento sencillo y directo para medir la actividad de la AST en una variedad de muestras. En este kit, la transferencia de un grupo amino del aspartato al α-cetoglutarato da como resultado la generación de glutamato, lo que da lugar a un producto colorimétrico (450 nm) proporcional a la actividad enzimática de AST presente. Una unidad de AST es la cantidad de enzima que generará 1,0 μmol de glutamato por minuto a pH 8,0 y a 37 °C.

  1. How is shipping temperature determined? And how is it related to the product storage temperature?

    1. Products may be shipped at a different temperature than the recommended long-term storage temperature. If the product quality is sensitive to short-term exposure to conditions other than the recommended long-term storage, it will be shipped on wet or dry-ice. If the product quality is NOT affected by short-term exposure to conditions other than the recommended long-term storage, it will be shipped at ambient temperature. As shipping routes are configured for minimum transit times, shipping at ambient temperature helps control shipping costs for our customers.   2. How can I determine the shelf life / expiration / retest date of this product?

    1. If this product has an expiration or retest date, it will be shown on the Certificate of Analysis (COA, CofA). If there is no retest or expiration date listed on the product's COA, we do not have suitable stability data to determine a shelf life. For these products, the only date on the COA will be the release date; a retest, expiration, or use-by-date will not be displayed.
      For all products, we recommend handling per defined conditions as printed in our product literature and website product descriptions. We recommend that products should be routinely inspected by customers to ensure they perform as expected.
      For products without retest or expiration dates, our standard warranty of 1 year from the date of shipment is applicable.

  3. Is it possible to use the MAK055 for an experiment that involves evaluating samples with and without B6 added? Additionally, will hemoglobin interfere if a blood hemolysate sample is used when assayed with this kit?

    1. The specific experiment in question has not been performed in the lab, and there are no references for the Aspartate Aminotransferase Activity Assay Kit where a similar experiment has been reported. Considering the nature of the assay and the fact that B6 is a co-enzyme of the ASP, necessary for its activity, in principle, the kit will be suitable to evaluate samples treated with B6. It will be necessary to evaluate any possible interferences and optimize user-specific conditions.
      It is possible that hemoglobin will interfere with the assay if hemolyzed blood is tested. Blood is generally not recommended as a sample, unless specifically mentioned in the assay, due to the possible interference as mentioned. Furthermore, the kit has not been tested with blood samples.

  4. Does the AST Activity Assay Kit (MAK055), which includes a positive control (MAK055F), have a specific species origin? For instance, is it Human AST?

  5. Do you anticipate phenol red in culture medium having an impact on the colorimetric read out at 450 nm? (I'm thinking there may be some spectral overlap)

    1. Ideally, culture media should be phenol red-free.
