General description
Los nucleótidos son moléculas orgánicas que actúan como los monómeros, o subunidades, de los ácidos nucleicos (como el ADN y el ARN). Las unidades estructurales de los ácidos nucleicos, los nucleótidos, consisten en una base nitrogenada (purina o pirimidina), un azúcar de cinco carbonos (ribosa o desoxirribosa) y por lo menos un grupo fosfato. Un nucleósido con un grupo fosfato da lugar a un nucleótido. La mezcla de desoxinucleótidos es una práctica disolución de dNTP premezclados que contiene las sales de sodio ultrapuras de dATP, dCTP, dGTP y TTP, 10 mM cada una, en agua para biología molecular de gran calidad. Un µl es suficiente para una reacción PCR estándar de 50 µl.
Application
La mezcla de dNTP se ha utilizado:
- en la amplificación mediante PCR del ADN genómico aislado de insectos, hongos[1], virus y humanos,[2]
- en la transcripción inversa del ARN total a ADNc.[3][3]
- como componente de la mezcla de amplificación del ADN para la reacción en cadena de la polimerasa (PCR)[1][4]
- PCR habitual y larga
- secuenciación manual y automatizada del ADN
- síntesis de ADNc[5] y reacciones de marcaje
