Application

Bradykinin acetate salt has been used:
  • to stimulate the contraction of guinea pig ileum
  • as a contractile agent in fibroblasts
  • In the stimulation of endothelium of human subcutaneous artery
  • To evoke Ca2+ signals in neuroblastoma-glioma hybrid cell line, NG108-15

Biochem/physiol Actions

Activates pain receptors; induces smooth muscle contraction; activates Ca2+-dependent nitric oxide synthetase; increases capillary permeability.
Bradykinin is a pro-inflammatory peptide that acts through G-protein-coupled receptors. It activates sphingolipid metabolism. Fibroblasts treated with bradykinin produce a rapid and important increase in ceramide which is followed by a transient rise in sphingosine content. The peptide also increases ceramide cellular content by rapidly mobilizing neutral glycolipids. Bradykinin plays an important role in the regulation of fluid and electrolyte balance, smooth muscle contraction, vasodilation and capillary permeability. The bradykinin potentiators, which possess no bradykinin activity themselves, enhance the activity of bradykinin.

The modulation of neuronal adenylylcyclase by Ca2+, acting via calmodulin, is a long-established example of a positive interaction between the Ca2(+)-mobilizing and cAMP-generating systems. Preguntas

  1. How should a solution of bradykinin acetate salt in 0.1M acetic acid be stored? Can it be further diluted in an aqueous buffer?

    1 respuesta
    1. Following reconstitution at 25 mg/mL in 0.1 M Acetic acid, aliquot and freeze at -20°C. Stock solutions are stable for up to 3 months at -20°C. The stability of dilute solutions has not been determined, however, Bradykinin fragment 1 - 7, (B4181) is part of a set of standards for Maldi-MS calibration (MSCAL1). In this application, the solution is diluted to 10 pmol/uL and stored at -20°C for 1 month. One may infer that dilute solutions of the whole molecule would be comparably stable when diluted and stored frozen.
