Descripción general

Vanadyl ribonucleoside complexes are low molecular weight inhibitors of ribonucleases.[1]

Aplicación

Vanadyl ribonucleoside complexes (VDR) are transition state analogs that bind to active sites of many RNases, inhibiting their activity. Because the RNases are not covalently modified by the complexes, VDR must be used at every stage of RNA extraction and purification. However, VDR also inhibit RNA polymerases and in vitro translation and therefore must be removed from final preparation of RNA.
Vanadyl ribonucleoside complexes has been used in hybridization mixture for RNA in situ hybridization.[2][3][4]

Acciones bioquímicas o fisiológicas

The vanadyl ribonucleoside complex has the ability to block the formation of ribosomal subunit in Staphylococcus aureus. The complex exhibits anti-microbial activity[1] These complexes also serve as powerful inhibitors of several enzymes with ribonucleolytic activity.[5]

    1. This product is a complex that has an estimated molecular weight of 422.5 g/mole. The solubility at 400 mM has not been tested. This product is offered in a prepared solution under product 94742 at a concentration of 200 mM. Based on the estimated molecular weight, a 0.4 M (400 mM) solution would have a concentration of 169 mg/mL. Solutions may be prepared in nuclease-free PBS containing 15% glycerol or 5% DMSO as a cryoprotectant. Solutions may then be stored in working aliquots at −20˚C. Avoid the use of EDTA in all buffer solutions as this will cause degradation of the complex.     1. The method to prepare a 200mM solution of the 94740 is proprietary and involves in-process testing. However, product 94742 and R3380 are prepared 200mM solutions of Vanadyl Ribonucleoside complexes.
