Descripción general

Iron, the most abundant transition metal in our bodies, is involved in a number of biologically important processes such as respiration, oxygen transport, and energy production in collaboration with oxygen. In living cells, iron exists mainly as ferrous (Fe2+) and ferric (Fe3+) ionic forms. As a major contributor to oxidative damage of cells, Fe2+ is implicated in serious diseases such as cancers and neurodegenerative disorders, because of its ability to produce harmful reactive oxygen species via contact with oxygen, superoxide, and hydrogen peroxide (H2O2).BioTracker far-red labile Fe2+ dye is a fluorescent probe that specifically detects labile iron (II) ions (Fe2+). This probe is designed to selectively react with only Fe2+ separately from other metal ions and irreversibly turns into a far-red fluorescent substance. It mainly localizes inside endoplasmic reticulum (ER) and/or golgi bodies. It can also be used with a flow cytometer equipped with a red laser. FerroFarRed shows almost no cytotoxicity at concentrations up to 100 μM (~20 times that of generally used concentrations). This product is same as FerroFarRedTM (GC903-01) from Goryo Chemical, Inc.Spectral Properties:Absorbance (max): 646nmEmission (max): 662nm (Far-red)Research Category:Live Cell Dye

Preguntas

1–3 de 3 Preguntas
  1. does this reagent stain Fe2+ localized only inside endoplasmic reticulum (ER)?

    1 respuesta
    1. This item localizes primarily, but not exclusively, inside the endoplasmic reticulum (ER). This compound may also localize in the Golgi. While not confirmed, there is some indication that the material may detect Fe2+ in the cytoplasmic pool or other organelles.

  2. Does the BioTracker FAr-red Labile Fe2+ Dye stain dead or non-viable tissue, or does it exclusively stain live cells as advertised?

    1 respuesta
    1. SCT037, BioTracker Far Red Labile Fe2+ Dye exclusively stains live or viable cells and does not have a staining protocol for use on dead or non-viable cells. SCT037 is equivalent to FerroFarRed™ from Goryo Chemical.

  3. Hi, I was wondering if the BioTracker Labile Iron dye # SCT037 can be freeze-thawed after being resuspended in DMSO for a second use. Also, is it fixable? Thank you.

    1 respuesta
    1. Storing as a solution is not recommended. In solution, the reagent degrades and the fluorescence background signal may increase.
Yes, the FerroFarRed can be fixed under mild conditions such as 3% Paraformaldehyde for 10min at 4 °C. The fluorescence of this dye significantly diminishes when fixed in 3% PFA for 20min at RT.
