General description

CENP-A is a 17 kDa histone H3- like protein.
Centromere protein A is a protein encoded by the CENPA gene in humans. It is referred as CenH3 and CENP-A. It is a histone H3 variant that has centromere-specific nucleosomal property and is located at neocentromere found on a chromosome 10. CENP-A- binding clusters is associated with nucleosomal blocks within the neocentromeric chromatin.

Immunogen

synthetic peptide corresponding to amino acids 32-49 located near the N-terminus of human CENP-A, conjugated to KLH.

Application

Anti-CENP-A antibody has been used in immunoblotting.
Anti-CENP-A antibody produced in rabbit is suitable for western blotting at a concentration of 1-2μg/mL using HEK-293 cells expressing human CENP-A.

Biochem/physiol Actions

The location of the centromere on the chromosome is dependent on the histone H3 variant centromere protein A (CENP-A). The N-terminal tail of CENP-A is highly different from other H3 variants. Serine 7 of (CENP-A) plays an important role in mitosis-specific phosphorylation. It also plays a key role in kinetochore assembly. Alteration in CENP-A causes various human cancers. CENP-A may play an important role in epithelial ovarian cancer (EOC) and may serve as prognostic marker. It acts as a potential target for gene therapy in the treatment of EOC. The NH2 terminus of human CENP-A is essential for mitotic progression and necessary for the platform function of CENP-A centromeric chromatin in the assembly as well as maintenance of active kinetochores.

Physical form

Solution in 0.01 M phosphate buffered saline, pH 7.4, containing 15 mM sodium azide.
