Application

La enzima se ha utilizado junto con la amiloglucosidasa para la digestión in vitro del almidón en muestras de alimento.[1] La pancreatina de Sigma se ha utilizado para ensayos de digestión pepsina-pancreatina in vitro durante la estimación de la digestibilidad del intestino delgado utilizando restos de alimentos del rumen. Se ha utilizado también junto con la pepsina para simular la digestión gástrica e ileal in vitro de materias primas en cerdos.
La pancreatina es una mezcla de diversas enzimas digestivas producidas por las células exocrinas del páncreas porcino. Es una proteasa de amplio espectro compuesta por amilasa, tripsina, lipasa, ribonucleasa y proteasa. Se utiliza para análisis de digestibilidad in vitro[2]. El producto P7545 se ha utilizado para analizar las sensibilidades de los inhibidores de bacterias celulolíticas[3].

Biochem/physiol Actions

Debido a los componentes enzimáticos como la tripsina, la amilasa y la lipasa, la pancreatina hidroliza proteínas, almidón y grasas. La pancreatina convertirá no menos de 25 veces su peso de almidón de patata en carbohidratos solubles en 5 minutos en agua a 40 °C, digerirá no menos de 25 veces su peso de caseína en 60 minutos a pH 7,5 a 40 °C y liberará no menos de 2 microequivalentes de ácido por minuto por miligramo de pancreatina del aceite de oliva a pH 9,0 y 37 °C.
La pancreatina contiene componentes enzimáticos, como la tripsina, la amilasa y las lipasas, las ribonucleasas y las proteasas, producidas por las células exocrinas de páncreas porcino. Esta combinación de enzimas permite hidrolizar las proteínas, el almidón y las grasas. La pancreatina convertirá no menos de 25 veces su peso de almidón de patata en carbohidratos solubles en 5 minutos en agua a 40 °C, digerirá no menos de 25 veces su peso de caseína en 60 minutos a pH 7,5 a 40 °C y liberará no menos de microequivalentes de ácido por minuto por miligramo de pancreatina del aceite de oliva a pH 9,0 y 37 °C.

  1. Can you please tell us regarding trypsin range (concentration) in the pancreatin. We get it very low about 0.8U/mg when performed assays, is it normal or it is indicated as low activity.

  3. Hello, I want to assess the effect of enzymes in pancreatin on fungus and fungal cell wall specially (with lipase), is there a recommended concentration to use for pancreatin solution? and what is the recommended dissolving solution in this case? Thanks

    1. This product has not been tested in-house for the application of assessing the effect of enzymes in pancreatin on fungus and fungal cell walls, specifically with lipase. A recommended concentration has not been established. Conducting a literature search may provide guidance on this matter.

    1. This product meets 8X the minimum USP specifications. According to the USP monograph for Pancreatin, the 1X minimum specification is 25 USP units amylase, 2 USP units lipase, and 25 USP units protease per milligram. The calculated protease activity for the 8X preparation then, would be at least 200 units per milligram. This is a calculated value based on the USP monograph and are not reported separately on the Certificate of Analysis.

  5. Hi, what is the dissovled soultion? It can't dissolved in water or 1 mM HCl

    1. This product is not tested for solubility. For assay purposes. it is dissolved at 40 mg/3mL PBS, pH 6.8. Trituration is required for 5 - 10 minutes. Historically, this product has also been dissolved in water at 20 mg/ml. with heating at 37 °C. This method yields a hazy solution.

  7. What is the solubility of this enzyme in water?

    1. This product is soluble in water at up to 20 mg/ml in water. Heat may be required. This concentration will yield a hazy solution.

    1. The specific activity of this pancreatin is specified to be at least 8 x the minimum USP standard.

      The USP defines a minimum specification of a 1 X Pancreatin as follows:
      - 25 USP protease units per mg.
      - 25 USP amylase units per mg.
      - 2 USP lipase units per mg.

      Therefore, activity will be 8 times that.
