Le bleu de trypan (TB), un colorant azoïque anionique hydrophile, ne peut traverser que la membrane des cellules mortes. Il permet ainsi de colorer en bleu les cellules et les tissus morts. Il est utilisé en microscopie quantitative dans une technique de coloration par exclusion de colorant pour distinguer les cellules de mammifères vivantes et mortes.
Le bleu de trypan (TB), un colorant azoïque anionique hydrophile, ne peut traverser que la membrane des cellules mortes. Il permet ainsi de colorer en bleu les cellules et les tissus morts. Il est utilisé en microscopie quantitative dans une technique de coloration par exclusion de colorant pour distinguer les cellules de mammifères vivantes et mortes.
Le bleu de trypan est un colorant servant à distinguer les cellules vivantes des cellules mortes. C′est un colorant vital qui n′est pas absorbé par les cellules viables en bonne santé, mais qui colore les cellules dont la membrane cellulaire est endommagée. De ce fait, seules les cellules mortes sont dénombrées. Cette méthode est parfois appelée méthode avec colorant d′exclusion.
Application
Cette solution de bleu de trypan a été utilisée :
- dans un essai de viabilité cellulaire pour dénombrer les cellules viables et les cellules mortes.
- dans un test de mobilisation ("rescue assay") pour dénombrer les cellules viables.
- dans une méthode de comptage par exclusion de bleu de trypan pour établir une courbe de prolifération.
- comme solution saline témoin et dans la préparation d'une solution de lipopolysaccharide (LPS) pour confirmer le succès d'une injection de LPS.
- dans un test d'exclusion de colorant avec du bleu de trypan pour déterminer la viabilité de cellules épithéliales gastriques (GEC).
- pour dénombrer des cellules GEC avec un hémocytomètre.

Why are there red fibers in Product T8154, Trypan blue solution?

A report of fibers in Product T8154, Trypan blue solution is sufficiently common almost to be a characteristic of the product. These fibers are a cosmetic fault rather than one of functionality. The presence of fibers may be more pronounced following cold storage or shipping. This product should be stored at room temperature. Filtration to remove the fibers will not affect the use of the product.

Trypan Blue has a greater affinity for serum proteins than for cellular protein. If the background is too dark, cells should be pelleted and resuspended in protein-free medium or salt solution prior to counting.

Trypan Blue is a vital stain recommended for use in estimating the proportion of viable cells in a population. The reactivity of this dye is based on the fact that the chromophore is negatively charged and does not react with the cell unless the membrane is damaged. Staining facilitates the visualization of cell morphology. Live (viable) cells do not take up the dye and dead (non-viable) cells do.
