General description

c-Myc-, N-Myc- and L-Myc-encoded proteins function in cell proliferation, differentiation and neoplastic disease. Myc proteins are nuclear proteins with relatively short half lives. Amplification of the c-Myc gene has been found in several types of human tumors including lung, breast and colon carcinomas, while the N-Myc gene has been found amplified in neuroblastomas. The L-Myc gene has been reported to be amplified and expressed at high level in human small cell lung carcinomas. The c-Myc protein is a transcription factor, which is encoded by the c-Myc gene on human chromosome 8q24. c-Myc is commonly activated in a variety of tumor cells and plays an important role in cellular proliferation, differentiation, apoptosis and cell cycle progression.

Specificity

Recognizes Myc isoforms Mr 50-67kDa.
Will also cross-react with avian and Xenopus.

Immunogen

Bacterially expressed fusion-protein corresponding to the full-length human Myc.

Application

Immunoprecipitation:
4 µg of a previous antibody immuno-precipitated Myc as shown by an independent laboratory.

Immunocytochemistry:
This antibody has been used for immunocytochemistry by an independent laboratory. (Ohtsuka, T., 1998)

Quality

Routinely evaluated by western blot on A431 lysates from EGF-stimulated human A431 cells, human A431 cells and HeLa nuclear extract.

Western Blot Analysis:
0.5-2 µg/mL of this lot detected Myc in A431 lysates from EGF-stimulated human A431 cells. 0.5-2 µg/mL of a previous lot detected Myc in RIPA lysates from human A431 cells and HeLa nuclear extract.

Purified rabbit polyclonal IgG in buffer containing PBS, 0.05% sodium azide with 2 mg/mL BSA. Frozen solution.

Storage and Stability

Stable for 1 year at -20°C from date of receipt.

Handling Recommendations:
Upon first thaw, and prior to removing the cap, centrifuge the vial and gently mix the solution. Aliquot into microcentrifuge tubes and store at -20°C. Avoid repeated freeze/thaw cycles, which may damage IgG and affect product performance.

Analysis Note

Control
Positive Antigen Control: Catalog #12-302, EGF-stimulated A431 cell lysate. Add 2.5µL of 2-mercaptoethanol/100µL of lysate and boil for 5 minutes to reduce the preparation. Load 20µg of reduced lysate per lane for minigels.
