Specificity

Clone 1C20 is a ZooMAb® Rabbit recombinant monoclonal antibody that specifically detects MCM7. It targets an epitope within 24 amino acids from the internal region.

Immunogen

KLH-conjugated linear peptide corresponding to 24 amino acids from the internal region of human MCM7.

Application

Quality Control Testing

Evaluated by Western Blotting in HEK293 cell lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected MCM7 in HEK293 cell lysate.

Tested Applications

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected MCM7 in COS-7, HeLa, NIH3T3, and C6 lysates.

Affinity Binding Assay: A representative lot of this antibody bound MCM7 peptide with a KD of 4.9 x 10-7 in an affinity binding assay.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected MCM7 in HeLa cells.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected MCM7 in Human tonsil tissue sections.

Target description

DNA replication licensing factor MCM7 (UniProt: P33993; also known as EC:3.6.4.12, CDC47 homolog, P1.1-MCM3) is encoded by the MCM7 (also known as CDC47, MCM2) gene (Gene ID: 4176) in human. Unwinding of parental DNA in eukaryotic cells is performed by the 11-subunit Cdc45 MCM GINS assembly. MCM7 is a component of a highly conserved, multiprotein helicase complex that is required for DNA replication and G1/S transition. The six MCM proteins, MCM2-7 that belong to the AAA+ family of ATPases form a toroidal hetero-hexameric ring that translocates on single-strand DNA. In the absence of DNA substrate, MCM2-7 adopt an open spiral conformation. This complex serves as the replicative helicase essential for ′once per cell cycle′ DNA replication initiation and elongation in eukaryotic cells. MCM proteins bind with chromatin before the initiation of DNA replication and are then released from the chromatin as DNA replication proceeds. The active ATPase sites in the MCM2-7 ring are formed through the interaction surfaces of two neighboring subunits such that a critical structure of a conserved arginine finger motif (aa 513-516 in MCM7) is provided in trans relative to the ATP-binding site of the Walker A box of the adjacent subunit. The MCM domain of MCM7 is localized in amino acids 332-538. Three isoforms of MCM7 have been described that are produced by alternative splicing. Higher expression of MCM7 is reported in hepatocellular carcinoma (HCC) and is associated with worse overall survival. Knockdown of MCM7 is shown to inhibit cellular proliferation and HCC tumorigenicity. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Reconstitution

0.03 mg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
