Anwendung

Quality Control Testing

Evaluated by Western Blotting with recombinant fragment of Human Trop2.

Western Blotting Analysis: A 1:10,000 dilution of this antibody detected His- and hFc-tagged recombinant fragment of Human Trop2.

Tested Applications

Flow Cytometry Analysis: 1 µg from a representative lot detected Trop2 in one million HaCaT cells.

Enzyme Immunoassay (ELISA) Analysis: A serial of dilutions from a representative lot detected His-and hFc-tagged recombinant fragment of Human Trop2.

Affinity Binding Assay: A representative lot of this antibody bound His and hFc-tagged recombinant fragment of Human Trop2 with a KD of 1.5 x 10-6 in an affinity binding assay.

Immunocytochemistry Analysis: A 1:100 filution from a representative lot detected Trop2 in HaCaT cells.

Zielbeschreibung

Tumor-associated calcium signal transducer 2 (UniProt: P09758; also known as Cell surface glycoprotein Trop-2, Trophoblast antigen 2, Membrane component chromosome 1 surface marker 1, Pancreatic carcinoma marker protein GA733-1) is encoded by the TACSTD2 (also known as GA733-1, M1S1, TROP2) gene (Gene ID: 4070) in human. Trop2 is a single-pass type 1 membrane glycoprotein that is synthesized with a signal peptide (aa 1-26), which is subsequently cleaved off to generate the mature form that contains an extracellular domain (aa 27-274), a transmembrane domain (aa 275-297), and a cytoplasmic domain (aa 298-323). Its expression is observed in placenta and is highly expressed in many cancers where it plays a role in tumor progression. About 85% of all solid cancers display increased expression of Trop2. Physikalische Form

Purified recombinant mouse monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Rekonstituierung

300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Lagerung und Haltbarkeit

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
