Der D-2-Hydroxyglutarat(D2HG)-Spiegel ist in normalen Zellen und Geweben niedrig, in Stoffwechselerkrankungen wie der seltenen autosomalen Störung D2HG-Azidurie jedoch deutlich erhöht. D2HG ist in anderen Stoffwechselerkrankungen, darunter dem multiplen Acyl-CoA-Dehydrogenase-Mangel, dem Dihydrolipoyl-Dehydrogenase-Mangel, dem Pyruvat-Decarboxylase-Mangel, dem Pyruvat-Carboxylase-Mangel, verschiedenen Krebserkrankungen sowie in Neoplasmen mit Mutationen in den Isocitrat-Dehydrogenase-1(IDH1)- und Isocitrat-Dehydrogenase-2(IDH2)-Genen leicht erhöht. Die Bestimmung eines erhöhten D2HG-Spiegels ist ein wichtiger Biomarker für die frühzeitige Diagnose, Prognose, Überwachung und Entwicklung von therapeutischen Strategien zur Bekämpfung dieser Krankheiten.

Eignung

Zur Bestimmung des D2HG-Spiegels in Zell- und Gewebelysaten, Serum, Urin, kultivierten Zellen und Kulturüberständen geeignet.

Prinzip

Das in Partnerschaft mit dem Deutschen Krebsforschungszentrum (DKFZ) entwickelte D2HG-Assay-Kit ist ein schneller und empfindlicher Enzym-Assay für die Bestimmung des D2HG-Spiegels in verschiedenen biologischen Flüssigkeiten: Serum, Urin, Zellkulturüberständen und Zell- oder Gewebelysaten. Der ursprünglich von Balss et al. entwickelte Assay basiert auf der Oxidation von D2HG zu α-Ketoglutarat (αKG) durch das Enzym (D)-2-Hydroxyglutarat-Dehydrogenase (HGDH), gekoppelt an die Reduktion von NAD+ zu NADH (siehe Abbildung 1). Die Menge des erzeugten NADH wird anschließend durch die diaphorasevermittelte Reduktion von Resazurin zum fluoreszierenden Farbstoff Resorufin quantifiziert (λex = 540 nm/λem = 590 nm).

  1. How is shipping temperature determined? And how is it related to the product storage temperature?

    1. Products may be shipped at a different temperature than the recommended long-term storage temperature. If the product quality is sensitive to short-term exposure to conditions other than the recommended long-term storage, it will be shipped on wet or dry-ice. If the product quality is NOT affected by short-term exposure to conditions other than the recommended long-term storage, it will be shipped at ambient temperature.   2. How can I determine the shelf life / expiration / retest date of this product?

    1. If this product has an expiration or retest date, it will be shown on the Certificate of Analysis (COA, CofA). If there is no retest or expiration date listed on the product's COA, we do not have suitable stability data to determine a shelf life. For these products, the only date on the COA will be the release date; a retest, expiration, or use-by-date will not be displayed.
      For all products, we recommend handling per defined conditions as printed in our product literature and website product descriptions. We recommend that products should be routinely inspected by customers to ensure they perform as expected.
      For products without retest or expiration dates, our standard warranty of 1 year from the date of shipment is applicable.

  3. What is the recommended approach for measuring 2-HG levels in cell culture samples using the MAK320 detection kit if the sample volume is very small (i.e. 100 ul) and there is no access to equipment for accurately measuring pH in small volumes? Additionally, is the pH range critical for the experiment to work? Would a higher or lower pH result in decreased enzyme activity or render the experiment ineffective?

    1. The recommended solution is to use litmus paper to measure the pH of the sample. Another option is to use a special electrode designed for very small volumes, although this can be complicated and expensive. While the pH of the sample is crucial for test performance, the pH range is wide, making pH paper suitable and requiring only a few microliters of the sample.
