Biochem./physiol. Wirkung

IRAK4 (interleukin-1 receptor-associated kinase 4) protein contains a death domain and a kinase-like domain. IRAKs are mainly involved in toll-like receptors (TLRs) and interleukin-1 receptors (IL-1Rs) signaling. IRAK4 can activate NF-κB (nuclear factor κ B) and MAPK (mitogen activated protein kinase) pathways. It plays a crucial role in adaptive immunity and inflammatory responses.[1][2][3] IRAK4 mRNA levels are higher in humans suffering from Vogt-Koyanagi-Harada (VKH) disease.[1] Mutations in IRAK4 are associated with recurrent pyogenic bacterial infections in children.[4]

Toll/IL-1 receptor family members are central components of host defense mechanisms in a variety of species. One well conserved element in their signal transduction is Ser/Thr kinases, which couple early signaling events in a receptor complex at the plasma membrane

Trans-autophosphorylation is among the most prevalent means of protein kinase activation, yet its molecular basis is poorly defined. In Toll-like receptor and interleukin-1 receptor signaling pathways, the kinase IRAK4 is recruited to the membrane-proximal adaptor MyD88 through death domain (DD)

IRAK4 is a central kinase in innate immunity, but the role of its kinase activity is controversial. The mechanism of activation for IRAK4 is currently unknown, and little is known about the role of IRAK4 kinase in cytokine production, particularly
