Streptavidin wird kovalent an Meerrettichperoxidase gekoppelt. Das Konjugat wird zum Nachweis von biotinmarkierten Substanzen verwendet.
Anwendung
Einheitendefinition
Eine Einheit Streptavidin-POD, die einer Einheit Peroxidase entspricht, oxidiert 1 μmol Guajacol-Tetramer bei +25 °C und einem pH von 7,0 in einer Minute.
Umrechnung der Einheiten: 1 U (+25 °C; Guajacol als Substrat; pH 7,0) entspricht 1,24 U (+25 °C; 1,7 mM ABTS als Substrat; pH = 6,0) oder 4,0 U (+25 °C; 8,7 mM ABTS als Substrat; pH = 5,0).
Umrechnung der Einheiten: 1 U (+25 °C; Guajacol als Substrat; pH 7,0) entspricht 1,24 U (+25 °C; 1,7 mM ABTS als Substrat; pH = 6,0) oder 4,0 U (+25 °C; 8,7 mM ABTS als Substrat; pH = 5,0).

Angaben zur Herstellung
Arbeitskonzentration: 5–500 mU/ml (d. h., Stammlösung kann verdünnt werden). Arbeitskonzentration des Konjugats ist abhängig von Anwendung und Substrat. Die folgenden Konzentrationen sind als Richtlinie zu verstehen:
- ELISA: 50 bis 200 mU/ml
- Immunhistozytochemie: 250 bis 1000 mU/ml
- Western Blot: 200 bis 500 mU/ml
