Specificity

Clone 2K11 is a Rabbit recombinant monoclonal antibody that specifically detects Cyclin A. It targets an epitope within 22 amino acids from the C-terminal region.

Application

Quality Control Testing

Evaluated by Western Blotting in Jurkat cell lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected Cyclin A2 in Jurkat cell lysate.

Tested applications

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected Cyclin A2 in U2OS, NIH3T3, and PC12 cell lysates.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected Cyclin A2 in human tonsil tissue sections.

Affinity Binding Assay: A representative lot of this antibody bound Cyclin A2 with a KD of 9.7 x 10-8 in an affinity binding assay.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected Cyclin A2 in U2OS cells.

Target description

Cyclin-A2 (UniProt: P20248; also known as Cyclin-A, Cyclin A) is encoded by the CCNA2 (also known as CCN1, CCNA) gene (Gene ID: 890) in human. Cyclins, as binding and activating partners of Cdk, are important regulatory components of the cell cycle machinery. In animal cells, both cyclin A and B play a key role in the control of mitosis. In mammalian cells two types of cyclin A have been described. Cyclin A1 is specifically expressed in the testis, and cyclin A2 is ubiquitously expressed. Cyclin A2 has also been linked to cell proliferation and is often found expressed at a high level in human cancers. Cyclin A2 controls both the G1/S and the G2/M transition phases of the cell cycle. It acts through the formation of specific serine/threonine protein kinase holoenzyme complexes with Cdk1 and Cdk2. The cyclin subunit confers the substrate specificity of these complexes and differentially interacts with and activates Cdk1 and Cdk2 throughout the cell cycle. Cyclin A2/Cdk activity initiates mitosis by phosphorylating and inactivating the protein kinase Wee1, resulting in activation and nuclear localization of cyclin B1/Cdk1. During cell-cycle progression, cyclin A2 level increases at the onset of S-phase and contributes to the stimulation of DNA synthesis. In early mitosis, it drives the initiation of chromosome condensation and nuclear envelope breakdown. Cyclin A2 is localized both in the nucleus and cytosol. It is exclusively nuclear during interphase and is detected in the nucleus and the cytoplasm at prophase. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Reconstitution

300 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
