Cholesterol was used to prepare immune-stimulating complexes to enhance cellular and humoral responses.[1] It was incorporated in animal feed to study the effect of cholesterol-rich diet.

Biochem/physiol Actions

Cholesterol is a lipid that makes up about 20-25% of the structural components of the cell membranes. It determines the fluidity and permeability of the membrane, making it permeable to water but not to ions and protons. Cholesterol also regulates the functions of the transporters and signaling proteins present on the plasma membrane.[2] The major sites of cholesterol synthesis are small intestine and liver.[3]
Major component of all biological membranes; ~25% of total brain lipid is cholesterol.

Preparation Note

Cholesterol monohydrate yields a clear, colorless to faint yellow solution in hot alcohol. It is also soluble in acetone, dioxane, ethyl acetate, benzene, petroleum ether, oils, fats, and in aqueous solutions of bile salts. Solutions should be protected from light.

  1. Hello, I would like to know how to dissolve this product in serum, and what its maximum solubility in serum is? Because I am looking for a cholesterol product to prepare a solution with a concentration >250 mg/dL. Thank you!

    1. Free cholesterol is practically insoluble in serum. A true solution at a concentration of 250 mg/dL cannot be reliably achieved. Typical cholesterol levels in human serum may range from 150 – 250 mg/dL, with 30 – 70 mg/dL as free cholesterol. The majority is bound to lipoproteins in the serum matrix. Higher concentrations may be possible with heat, however this has not been investigated. The end user would have to determine the rate. Please see the link below to review a water-soluble cholesterol formulation, C4951. This product is a cyclodextrin complex that enhances cholesterol solubility up to 300 mg/mL (approximately 50 mg cholesterol).

      For the Cell Culture Tested grade of this product, C3045, three options are provided: Serum, Tween and Ethanol.
      Note that if Serum is used as the solvent, it may already contain some Cholesterol. The "Tween" is referring to an aqueous solution of either TWEEN 20 or TWEEN 80. Also, Ethanol can be used to make a stock solution, before supplementing the culture medium; consider perhaps a 100X solution in ethanol, which would give a concentration of 1 percent ethanol in the medium.

  3. What is the best temperature for storage for 5 mg/mL solutions in ethanol? Can they be stored at 4C? Thank you!

    1. The stability of this material in solution at 2-8°C has not been evaluated. Solutions prepared in Chloroform, in aliquots and protected from light, are stable for at least 1 year when stored at -20°C. Solutions prepared in Ethanol are expected to be similarly stable.

  4. How can I determine the shelf life / expiration / retest date of this product?

    1. If this product has an expiration or retest date, it will be shown on the Certificate of Analysis (COA, CofA). If there is no retest or expiration date listed on the product's COA, we do not have suitable stability data to determine a shelf life. For these products, the only date on the COA will be the release date; a retest, expiration, or use-by-date will not be displayed.
      For all products, we recommend handling per defined conditions as printed in our product literature and website product descriptions. We recommend that products should be routinely inspected by customers to ensure they perform as expected.
      For products without retest or expiration dates, our standard warranty of 1 year from the date of shipment is applicable.

  6. How is shipping temperature determined? And how is it related to the product storage temperature?

    1. Products may be shipped at a different temperature than the recommended long-term storage temperature. If the product quality is sensitive to short-term exposure to conditions other than the recommended long-term storage, it will be shipped on wet or dry-ice. If the product quality is NOT affected by short-term exposure to conditions other than the recommended long-term storage, it will be shipped at ambient temperature. As shipping routes are configured for minimum transit times, shipping at ambient temperature helps control shipping costs for our customers.
