General description

PureCol collagen is approximately 97% Type I with the remainder being comprised of Type III collagen. It is soluble atelo-collagen. It contains high monomer content as measured by gel permeation chromatography.

It is ideal for coating of surfaces and providing preparations of thin layers for culturing cells. PureCol, lyophilized form is not recommended for the formation of a solid gel. PureCol collagen is provided in user-friendly packaging for use and storage. This product is supplied as a sterile lyophilized powder.

Application

PureCol has been used:
  • in collagen gel preparation for the 3D sprouting assay with human umbilical vein endothelial cells (HUVEC) spheroids[1] 
  • to coat Costar® Transwell® Inserts plates before cell migration assays with bladder smooth muscle cells (BMSC)[2]
  • to coat culture plates for collagen degradation assay with breast cancer cell lines[3]

Preparation Note

This product is supplied as a lyophilized powder with 15 mg of bovine collagen. When reconstituted with 5 ml of sterile 0.01 N HCl, a concentration of approximately 3 mg/ml is achieved.

  1. Hi, I am going to find bovine collagen standard (purified collagen) for using in MALDI TOF analysis. I wonder whether the collagen product 5006 is suitable in this regard. what is your other recommendation?

    1. This product is not suitable for use in MALDI TOF analysis. A bovine collagen standard that is suitable for MALDI TOF analysis is not available.

  2. How can this product be sterilised after reconstitution to 3 mg/ml?

    1. The product is supplied sterile and should be reconstituted with 5ml of sterile 0.01M HCl. We would recommend filter sterilising the 0.01M HCl without collagen using a Millex syringe filter such as SLLG025SS (solvent resistant PTFE) or as it is only a weak acid solution then you could use cheaper SLGVR33RS (PVDF) sterile filters. It is not advisable to filter the collagen once in solution.

  3. Howlong is this product stable after reconstructiond and under which conditions should this stock solution be stored?

    1. The long term stability of this product in solution has not been determined. However, historical data suggests that solutions stored protected from light, in amber glass or wrapped in foil, are stable at least 30 days when stored refrigerated at 2 - 8°C.
