Application

CHO-K1 cell line from hamster has been used to study the effect of plasmid pCM01 induction on the association of bacteria with CHO-K1 cells by the gentamicin protection assay. It has also been cultured with or without lactate to measure time-series for cell proliferation.

Biochem/physiol Actions

A subclone of the parental CHO cell line, which was derived from the ovary of an adult Chinese hamster. Cells require proline due to the absence of the gene for proline synthesis, the block in the biosynthetic chain lies in the step converting glutamic acid to glutamine gamma serialdehyde. They undergo morphological changes in response to cholera toxin.

Preparation Note

Ham′s F12+ 2mM Glutamine+ 10% Foetal Bovine Serum FBS / FCS.
Split sub-confluent cultures (70-80%) 1:4 to 1:10 i.e. seeding at 1-2x10,000 cells/cm2 using 0.25%Trypsin or Trypsin/EDTA; 5% CO2; 37°C.

  2. Is the cell like deficient of DHFR gene?

    1. CHO-K1 are dihydrofolate reductase (DHFR) positive. There are other mutants made from CHO-K1 that are DHFR deficient. Please see the links below to review these possible options:

  5. What passage are my Cell Line cells?

    1. If the passage of the cells is known, it is listed on the product page at the HPA website (www.hpacultures.org.uk).

  6. How many Cell Line cells are in the vial?

    1 answer
    1. Each vial of cells contains 2-3 x 106 cells in 1 ml of freezing media. This is in a 1.8 mL ampule.  For exceptionally large cells, counts may decrease. Suspension cells, generally smaller cells, may contain as many as 4-5 x 106 cells/vial to assure optimal viability upon thaw.

  7. How do I handle Cell Line frozen cells upon arrival?

    1 answer
    1. Upon receipt, frozen ampules should be transferred directly to vapor phase liquid nitrogen without delay (-135°C) or liquid phase liquid nitrogen if vapor phase is not available. DO NOT use a -80°C freezer as an alternative; this will result in loss of viability.

  8. Why is Vapor Phase Liquid Nitrogen preferred for storage of Cell Line cells?

    1 answer
    1. If ampules are immersed into liquid phase of liquid nitrogen, it increases the risk of the liquid seeping into the vial. This could lead to problems of cross-contamination and increased risk of the ampule exploding when thawed.

  9. What medium should I use when I thaw the Cell Line cells?

    1 answer
    1. The medium for each cell line is listed on the product page. It is specific to each cell line. If not visible on the Sigma-Aldrich website product page, the HPA website also contains the same information (www.hpacultures.org.uk). The product number for the cell line is the same on either site.
