General description

LIMD1 (LIM domains-containing protein 1) is a tumor suppressor and member of the Ajuba family of LIM domain-containing proteins. LIMD1 interacts with retinoblastoma protein (pRB), inhibits E2F-mediated transcription, and suppresses the expression of the majority of genes with E2F1-responsive elements. Recently, LIMD1 has been implicated in regulating cellular processes involved in hypoxia. LIMD1 acts as a molecular scaffold, simultaneously binding the prolyl hydroxylases (PHDs) and von Hippel Lindau tumor suppressors (VHL), thereby assembling a PHD-LIMD1-VHL protein complex and creating an enzymatic niche that enables efficient degradation of HIF-1α.
