General description

DNA polymerase beta (EC 2.7.7.7; UniProt P06746; also known as DNA pol beta, DNA polymerase beta subunit) is encoded by the POLB gene (Gene ID 5423) in human. DNA polymerase beta is involved in DNA recombination and base-excision repair (BER). Upon oxidative DNA damage, Polb translocates to the nucleus, where it associates with AP endonuclease 1 (Apex) to carry out DNA repair. Apex makes a single nick 5′ to an abasic site in double-stranded DNA, Polb then inserts the replacement nucleotide(s). Aberrant POLB overexpression results in an increased rate of spontaneous mutagenesis and a highly mutagenic tolerance phenotype. Thirty percent of all tumors are reported to harbor mutations in the POLB gene.

Application

Immunohistochemistry Analysis: A 1:50-250 dilution from a representative lot detected DNA polymerase beta in normal rat testis, normal human kidney, as well as human lung & prostate cancer tissue sections.
This Anti-DNA polymerase beta Antibody is validated for use in Western Blotting, Immunohistochemistry for the detection of DNA polymerase beta.

Analysis Note

Evaluated by Western Blotting in human testis tissue lysate.

Western Blotting Analysis: 1.0 µg/mL of this antibody detected DNA polymerase beta in 10 µg of human and mouse testis tissue, Jurkat, and HeLa lysates.

A major focus of breast cancer research is to understand the mechanisms responsible for disease progression and drug resistance. Toward that end, it has been found that approximately two thirds of all human breast carcinomas overexpress the Estrogen Receptor α (ERα) protein and it remains the primary pharmacological target for endocrine therapy1,2. The normal cellular function of ERα is as a transcription factor that mediates a wide variety of physiological processes, many of which are dependent upon phosphorylation of the receptor at specific amino acid residues3,4. Indeed, ERα is known to be phosphorylated at a multitude of different sites, yet how these all correlate to disease remains unclear5. Here, we interrogated multiple sites of ERα for phosphorylation status by screening an extensive panel of different breast cancer patient samples and other non-breast cancer tissue microarray (TMA) slide samples to determine their relevance to disease.
