General description
La glutatión reductasa (GLR1) existe en las isoformas mitocondrial y citoplásmica. Comparte secuencia y homología estructural con la tiorredoxina reductasa, y es una oxidorreductasa que contiene flavina. Su sitio activo se compone de un disulfuro rédox activo , y requiere NADPH para su actividad catalítica. Es una enzima ampliamente presente y se encuentra en plantas, bacterias, levaduras, ratones y seres humanos.[1]

La glutatión reductasa (GR) de la levadura de panadero′( Saccharomyces cerevisiae) se ha utilizado-
- para cuantificar el contenido de glutatión en el tejido miocárdico utilizando un ensayo de reciclado enzimático basado en la glutatión reductasa-5,5′-ditiobis (ácido 2-nitrobenzoico)[3]
- para la cuantificación de glutatión reducido (GSH) en los ovocitos, utilizando un ensayo de microglutatión ligeramente modificado, obtenido de doradas prepuberales[2]
- para la preparación de GSSG (disulfuro de glutatión) total + medición de GSH, donde todos los GSSG disponibles se redujeron a GSH, en cristalino de rata[6]
- para la cuantificación del glutatión reducido intracelular (GSH) en los ovocitos obtenidos de ratas[7]
Biochem/physiol Actions
El glutatión (γ-glutamilcisteinilglicina) es un tripéptido tiol ubiquo que desempeña un papel crucial en el mecanismo de defensa frente al estrés oxidativo de la célula. La glutatión reductasa (GLR1) es responsable de la reducción del disulfuro de glutatión (GSSG) a glutatión reducido (GSH).[1]
La glutatión reductasa (IGR) es una flavoenzima crucial en el sistema de defensa antioxidante. La glutatión peroxidasa utiliza el glutatión reducido (GSH) para detoxificar el peróxido de hidrógeno y en el proceso lo convierte en glutatión oxidado (GSSG). El GSSG es reciclado de nuevo a GSH por la glutatión reductasa (GR) utilizando NADPH que se convierte entonces en NADP+. El GSH regenerado está entonces disponible para detoxificar más peróxido de hidrógeno. La enzima utiliza FAD como cofactor. La GR y la glutatión peroxidasa pueden inhibir la peroxidación lipídica actuando como enzimas antioxidantes en el esperma. La glutatión reductasa comparte un motivo estructural con una serie de otras proteínas, como las aspartil proteasas, la citrato sintasa, las manos EF, las hemoglobinas, las lipocalinas y las α/β hidrolasas. The volume can be determined using the activity value reported on the lot specific Certificate of Analysis.
For example, if the activity is reported to be 224 units/mg protein (Biuret):
This equates to 5.2 units /mL protein (Biuret)100 units / 224 units/mg =0.446mg, that is, 100 units = 0.446 mg protein
If the protein content is 5.2 mg/ml, then 0.446mg should be approximately 0.086 ml=86 μLHelpful?
