General description
El sulfato de bleomicina es un antibiótico antineoplásico aislado de Streptomyces verticillus. Se recomienda su uso como agente de cultivo celular a 10-100μg/ ml.

Application
La bleomicina se utiliza en combinación con etopósido y cisplatino para tratar el cáncer testicular, de ovarios y de cuello uterino. Esta combinación se ha hecho popular como quimioterapia BEP y se ha demostrado como el régimen quimioterápico eficaz y más satisfactorio en los tumores de células germinales. Dado que es un citotóxico, se sabe que la bleomicina induce lesión endotelial en la vasculatura pulmonar lo que se conoce como lesión pulmonar inducida por bleomicina o neumonitis inducida por bleomicina (BIP). Los investigadores han utilizado esta propiedad de la bleomicina para inducir fibrosis pulmonar en modelos de rata y ratón.
Biochem/physiol Actions
El sulfato de bleomicina se une al ADN, provoca la escisión del ADN monocatenario en secuencias de bases específicas e inhibe la síntesis de ADN. Esta acción inhibidora requiere que la bleomicina se una al oxígeno y a un ion metálico. También puede escindir el ARN en menor grado pero con más selectividad. Actúa como inductor y regulador de la apoptosis e inhibe la angiogénesis tumoral.
Components
Este producto es una mezcla de antibióticos glucopeptídicos que contienen bleomicina A2 (70 %) y B2(30 %). Las bleomicinas difieren entre sí en la amina terminal y muestran actividad biológica variable.
Caution
Este producto debe conservarse a 2-8°C.
Preparation Note
El producto es soluble en agua a 20 mg/ml y es activo durante varios días. Una disolución de 2 unidades/ml en fosfato de potasio neutro 0,1 M debe utilizarse en 14 días. Una disolución es activa durante tres meses a 3 unidades/ml en disolución salina normal.
