General description

Manganese superoxide dismutase (MnSOD) protein is a homotetrameric SOD. This protein is composed of four subunits, each containing one Mn2+. MnSOD enzyme is synthesized in the cytosol and is modified for its transport into the mitochondria where it resides in the matrix.
The SOD2 (superoxide dismutase 2) gene is mapped to human chromosome 6q25.3. The encoded protein functions as a tetramer and is localized to mitochondria.

Specificity

Anti-Superoxide Dismutase (MnSOD) (KC-19) recognizes human and rat Superoxide Dismutase (MnSOD).

Immunogen

synthetic peptide corresponding to amino acid residues 202-220 of human superoxide dismutase (MnSOD), conjugated to KLH. The corresponding sequence is identical in apes and monkeys, differs by one amino acid in mouse and horse, by two amino acids in cow, and by four amino acids in rat.

Application

Anti-Superoxide Dismutase (MnSOD) (KC-19) antibody produced in rabbit may be used in immunoblotting and indirect immunofluorescence.

Biochem/physiol Actions

Manganese superoxide dismutase (MnSOD) is essential for the survival of aerobic life. This protein is inducible by tumor necrosis factor (TNF) and protects cells from TNF-mediated apoptosis. MnSOD is proposed to function as a tumor suppressor by modulating the activity of redox-sensitive transcription factors and specific signal mediators. Overexpression of the MnSOD gene protects neurons from N-methyl-D-aspartate receptor (NMDA) and nitric oxide-induced neurotoxicity. A decline in the MnSOD activity occurs in aging, progeria, cancer, asthma and transplant rejection.
Superoxide Dismutase 2 (SOD2) is an antioxidant enzyme and catalyzes the conversion of superoxide to hydrogen peroxide and molecular oxygen. The enzyme offers protection against oxidative damage induced by reactive oxygen species and also is radioprotective. The gene is associated with the metastasis and invasion process of salivary adenoid cystic carcinoma. Manganese SOD is involved in detoxification.

Physical form

Solution in phosphate buffered saline, pH 7.4, containing 15 mM sodium azide.

Storage and Stability

For continuous use, store at 2-8 °C for up to one month. For extended storage, freeze in working aliquots. Repeated freezing and thawing is not recommended. Storage in frost-free freezers is also not recommended. If slight turbidity occurs upon prolonged storage, clarify the solution by centrifugation before use. Working dilutions should be discarded if not used within 12 hours.
