General description

Sirtuin 3 (Sirt3) is an NAD-dependent deacetylase localized to the mitochondrial matrix. SIRT3 encodes a 44 kDa full length form and a N-terminal 142 amino acids truncated form. It is mapped to human chromosome 11p15.5.

Immunogen

synthetic peptide corresponding to amino acids 384-399 of human SIRT3, conjugated to KLH via an N-terminal cysteine residue.

Application

Anti-Sirt3 antibody produced in rabbit has been used in western blotting.

Biochem/physiol Actions

Sirtuin 3 (Sirt3) is mainly a nuclear protein that transfers to the mitochondria during cellular stress. In humans, Sirt3 deacetylates and activates the mitochondrial enzyme acetyl-CoA synthetase 1 (AceCS2). Sirt3 is implicated in regulating mitochondrial function and thermogenesis in brown adipocytes. Its levels have been shown to correlate with extended life span. Increased levels of Sirt3 were associated with node positive breast cancer.

Physical form

Solution in 0.01 M phosphate buffered saline, pH 7.4, containing 15 mM sodium azide.
