General description

Application

Pectolyase P5936 (Pel1) is a natural mixed pectolyase produced by the fungus Aspergillus japonicus used to digest components (endopolygalacturonate and pectin) of plant cell walls. Treatment of cell walls with pectolyase can be used to destablilize the cell walls for organelle isolation or to modify the elasticity of the cell walls.
Pectolyase from Aspergillus japonicas has been used to digest the siliques and also in immunofluorescence experiments.
Used in plant protoplast preparation to digest cell wall prior to organelle isolation.

Other Notes

Reported to contain two types of pectinase, endopolygalacturonase (EC3.2.1.15), endopectin lyase (EC4.2.2.10) and a maceration stimulating factor.

Unit Definition

One unit will liberate 1.0 μmole of galacturonic acid from polygalacturonic acid per min at pH 5.5 at 25 °C.
