Descripción general

Neutrophil Elastase (NE, EC 3.4.21.37, leukocyte elastase, ELANE, ELA2, elastase 2, neutrophil elaszym, or serine elastase) is a cytotoxic serine protease. It is stored in the azurophil granules of neutrophil granulocyte and is released following cell stimulation, e.g. by pathogens, immune complexes or chemotactic agents (PMA). Mutation in the gene ELA2 (neutrophil elastase) results in neutropenia, a hereditary disorder characterized by reduced neutrophil count in an individual.[1]

Principio

The Neutrophil Elastase Activity Assay Kit utilizes the ability of NE to proteolytically cleave a synthetic substrate and release a fluorophore, AFC, which can be easily quantified by fluorescence. This assay kit is simple, rapid, and can detect as low as 1 ng of neutrophil elastase in a variety of samples.

    1. Products may be shipped at a different temperature than the recommended long-term storage temperature. If the product quality is sensitive to short-term exposure to conditions other than the recommended long-term storage, it will be shipped on wet or dry-ice. If the product quality is NOT affected by short-term exposure to conditions other than the recommended long-term storage, it will be shipped at ambient temperature.   2. How can I determine the shelf life / expiration / retest date of this product?

    1. If this product has an expiration or retest date, it will be shown on the Certificate of Analysis (COA, CofA). If there is no retest or expiration date listed on the product's COA, we do not have suitable stability data to determine a shelf life. For these products, the only date on the COA will be the release date; a retest, expiration, or use-by-date will not be displayed.
      For all products, we recommend handling per defined conditions as printed in our product literature and website product descriptions. We recommend that products should be routinely inspected by customers to ensure they perform as expected.
      For products without retest or expiration dates, our standard warranty of 1 year from the date of shipment is applicable.

    1. This kit can be used to determine neutrophil elastase activity from both species. Please review the article citations below where researchers have reported this kit being used with neutrophils isolated from human blood and mice brain.

  4. Hi I am wondering if you have any references where this assay has been used with Human serum. Please let me know. Thanks.

    1. A detailed search of the scientific literature does not reveal any references to utilizing this kit with human serum. This kit has not been tested directly on serum samples. In theory, it could possibly work as long as the serum sample has active elastase. This may require some research and development to utilize the kit for the intended purpose. Please also keep in mind that the kit is used primarily for elastase from neutrophils which is a type of white blood cell. The serum should be devoid of white blood cells from the collection process.
