General description

Monoclonal Anti-Human Macrophages (mouse IgM isotype) is derived from the LN-5 hybridoma produced by the fusion of mouse myeloma cells and splenocytes from immunized BALB/c mice. Macrophages are widely distributed throughout the body. They belong to the mononuclear phagocytes family. Macrophages have abundant cytoplasm and are also termed histiocyte.

Immunogen

deparaffinized human peripheral blood mononuclear cells

Biochem/physiol Actions

Macrophages play a crucial role in body defense mechanisms and have phagocytic functionality. They are heterogeneous in morphology, cytochemistry, function and surface antigens. Monoclonal antibodies to macrophage and histiocyte surface or cytoplasmic antigens are important for the immunodiagnosis of both benign and malignant histiocytic lesions. They are useful in the investigation of macrophage differentiation and maturation and the diagnosis of various inflammation.

Physical form

The product is provided as ascites fluid with 0.1% sodium azide as a preservative.

Storage and Stability

For continuous use, store at 2-8 °C for up to one month. For extended storage freeze in working aliquots. Repeated freezing and thawing is not recommended. Storage in "frost-free" freezers is not recommended. If slight turbidity occurs upon prolonged storage, clarify the solution by centrifugation before use.

Macrophages are distributed in tissues throughout the body and contribute to both homeostasis and disease. Recently, it has become evident that most adult tissue macrophages originate during embryonic development and not from circulating monocytes. Each tissue has its own composition

Benign and malignant proliferations of histiocytes and dendritic cells may be encountered in lymph nodes. Reactive histiocytic and dendritic cell infiltrates occur in response to diverse stimuli and in addition to causing lymphadenopathy, may be present unexpectedly in lymph nodes

Macrophages are effector cells of the innate immune system that phagocytose bacteria and secrete both pro-inflammatory and antimicrobial mediators. In addition, macrophages play an important role in eliminating diseased and damaged cells through their programmed cell death. Generally, macrophages ingest

Macrophages are present in all vertebrate tissues, from mid-gestation throughout life, constituting a widely dispersed organ system. They promote homeostasis by responding to internal and external changes within the body, not only as phagocytes in defence against microbes and in
