General description

E3 ubiquitin-protein ligase UBR5 (UniProt: O95071; also known as EC:2.3.2.26, E3 ubiquitin-protein ligase, HECT domain-containing 1, HECT-type E3 ubiquitin transferase UBR5, Hyperplastic discs protein homolog, hHYD, Progestin-induced protein) is encoded by the UBR5 (also known as EDD, EDD1, HYD, KIAA0896) gene (Gene ID: 51366) in human. UBR5 is a nuclear protein that is widely expressed and is shown to be most abundant in testis and is expressed at high levels in brain, pituitary, and kidney. Two isoforms of UBR5 are described that are produced by alternative splicing and differ only in a single amino acid. It is identified as a novel regulator of p53 and downregulation of UBR5 results in elevated levels of p53 in normal and transformed cells. Its deficiency in mice is reported to result in mid gestation embryonic death due to a compromised yolk sac vascular development. UBR5 is a component of the N-end rule pathway. It recognizes and binds to proteins bearing specific N-terminal residues that are destabilizing according to the N-end rule, leading to their ubiquitination and subsequent degradation. It is shown to be involved in maturation and/or transcriptional regulation of mRNA by activating CDK9 by polyubiquitination and is believed to play a role in control of cell cycle progression. It associates with CDK9 and TFIIS/TCEA1 and forms a transcription regulatory complex made of CDK9, RNAP II, UBR5, and TFIIS/TCEA1 that can stimulate target gene transcription. UBR5 regulates DNA topoisomerase II binding protein (TopBP1) in the DNA damage response and may have a role in tumor suppression. (Ref.: Smits, VAJ (2012). Cell Cycle 11(4); 715-720).

Storage and Stability

Stable for 1 year at -20°C from date of receipt. Handling Recommendations: Upon receipt and prior to removing the cap, centrifuge the vial and gently mix the solution. Aliquot into microcentrifuge tubes and store at -20°C. Avoid repeated freeze/thaw cycles, which may damage IgG and affect product performance.
