General description

La tirosinasa es una oxidasa que contiene cobre con actividad para catecoles y cresol. La tirosinasa participa en la conversión de la tirosinasa a dihidroxifenilalanina (DOPA). La polimerización de la DOPA lleva a la formación de melanina. La tirosinasa desempeña un papel en la reacciones del bronceado. Se dice que la enzima tiene dos sitios de unión para sustratos aromáticos y un sitio de unión aparte para oxígeno-cobre.

El pH óptimo es 6-7

Peso molecular: 128 kDa por difusión de velocidad de sedimentación; 133 kDa mediante medición de dispersión de la luz y 119,5 kDa por electroforesis.

La tirosinasa es una oxidasa que contiene cobre, con actividad para catecoles y cresol. Es responsable de las reacciones de pardeamiento. Se ha comunicado que la enzima tiene dos sitios de unión para sustratos aromáticos y un sitio de unión diferente para oxígeno-cobre. El cobre es probablemente el Cu(I), y su inactivación implicará la oxidación a Cu(II).

Application

La actividad tirosinasa se ha evaluado en un estudio en el que se desarrollaba un agente terapéutico alternativo para el tratamiento de la hiperpigmentación. [1] La tirosinasa también se ha utilizado en un estudio en el que se investigaba el fenotipo de albinismo oculocutáneo en la población paquistaní. [2]

  1. Is it possible to use an equivalent buffer, such as phosphate buffer (Na/K), instead of the recommended potassium phosphate buffer to reconstitute the enzyme?

    1. Tyrosinase is soluble (2 mg/mL) in 50 mM potassium phosphate buffer, pH 6.5, yielding a clear brown solution.
      The use of other buffers, such as phosphate buffer has not been evaluated. For optimal results and to minimize the risk of issues, we recommend closely following the provided instructions.

  2. Is it possible to reconstitute T3824-25KU with a solution other than potassium phosphate buffer?

    1. The enzymatic assays are performed using potassium phosphate buffer as a solvent. Unfortunately, no other solvent was tested in-house. An online search indicates that water may be used to initially reconstitute the enzyme and then further dilute to the solution suitable for the application.

    1. This product is not food grade. Unless otherwise stated, this product is intended for research use only. It is not suitable for any other purpose, which includes, but is not limited to, unauthorized commercial use, in vitro diagnostic use, or therapeutic use in humans or animals. Currently, a food grade product option is not available.

    1. This product is sold by activity. The minimum activity specification for this material is 1000 units per milligram. The weight of this product can be determined using the activity value reported on the lot specific Certificate of Analysis(COA). For example, the sample COA has a activity value of 6540 units/mg. The amount of powder in the 25,000 unit package will be 3.82mg.

  10. I see that the approximate activity of Product T3824, Tyrosinase from mushroom, is  ?1000 unit/mg solid, but how can I find the lot specific activity of this tyrosinase?

    1. The certificate of analysis for the lot will report the exact tyrosinase activity for a particular batch. The lot number can be found on the label of the bottle, or you can call technical service (800-325-5832) for a sample lot number to view the online certificate of analysis.
