General description

La mucina está compuesta principalmente por unidades de carbohidratos, cruciales para la arquitectura estructural y la conformación. El monómero de mucina pesa 640 kDa y la proteína contribuye sólo al 20 % del peso molecular. El componente carbohidrato está unido a las cadenas laterales de serina y treonina. Las mucinas se clasifican en mucinas secretadas y mucinas transmembranarias.
Las mucinas o glucoproteínas del moco son los principales componentes macromoleculares del moco de mamíferos.[1] Se utilizó también en un estudio para evaluar los frotis de esputo concentrados por cito-centrifugación para la detección de bacilos acidorresistentes.[2]

Application

La mucina de estómago porcino se ha utilizado:
  • en la generación de una curva patrón para la cuantificación de la mucina-2 generada durante la inflamación de las mucosas en ratones
  • como componente de los líquidos de la digestión en modelos de digestión humana in vitro[3]
  • como componente de saliva simulada

Se utilizó mucina de estómago porcino en estudios sobre el sitio de unión del PA-IL aglutinina específico de galactosa de Pseudomonas aeruginosa.[4]

Biochem/physiol Actions

La mucina tiene amplias aplicaciones farmacológicas en la administración de medicamentos. Desempeñan un papel protector en la inflamación. La desregulación en la síntesis de mucinas contribuye a la tumorogénesis y a la inflamación crónica.
El moco forma la cubierta protectora de todas las superficies epiteliales. El principal componente estructural del moco son las mucinas formadoras de gel La capa de moco que cubre el epitelio intestinal tiene la mucina MUC2 como molécula central. La MUC2 es producida por las células caliciformes.[5]

    1. The solubility in water has not been evaluated for these products. It is recommended to follow the reconstitution instructions and reconstitute at a concentration of up to 20 mg/mL in 1 M NaOH.

  2. What is the best way to sterilise the product without destroying the structure?

    1. The structural integrity of this product has not been evaluated post-sterilization. A general procedure for sterilizing mucin has been published as follows:
      1. Place dry powdered mucin in a flask.
      2. Cover the mucin with 95% ethanol.
      3. Heat the covered mucin at 70 °C for 24 hours.
      4. Evaporate the alcohol to recover the mucin.

  3. How does this differ from human mucin?

    1. This product has not been tested in comparison to human-source mucin.

  4. I saw that this product is made by peptide degradation. by that way, can it harvest transmembrane mucins? I wonder this product contains transmembrane mucin too, especially MUC1.

    1. This product is a very crude preparation of mucin obtained by digestion of hog stomach with pepsin, followed by precipitation of the mucin with alcohol. The crude mucin is collected by filtration and dried. It is a mixture of glycoproteins. Transmembrane mucins tend to be heavily glycosylated and are more resistant to digestion however, conditions such as pH, temperature, and activity of the enzyme may lead to degradation. Further characterization of the mucins present in the final product is not performed.

    1. This product is a crude mixture of glycoproteins. The sugar and amino acid composition of the mucin has not been determined.

    1. This crude mucin may not be readily soluble in solution. At 20 mg/mL in 1 M sodium hydroxide, turbid suspensions are observed. To try to increase the solubility, one might try further dilution and sonication, but we have not tried this in our labs.

    1. Product No. M2378 is a crude mucin preparation. Product No. M1778 is a partially purified powder prepared according to the method described in Glenister, D.A. and Salmon. K. Microbial Ecol. in Health & Disease 1, 31, (1988). The amount of bound sialic acids are also different. Please refer to the specification sheets on our website.
