General description

La formalina tamponada neutra (NBF) al 10% es el fijador utilizado con más frecuencia en todo el mundo para el microscopio óptico y es un fijador algo indulgente. La NBF al 10% es idónea para laboratorios de gran rendimiento y requiere un periodo de fijación relativamente corto, pero también puede utilizarse para el almacenamiento prolongado de tejidos. Se cree que la fijación con formalina se forma entre los aldehídos y las proteínas, creando un gel y, por tanto, reteniendo los constituyentes celulares en su relación in vivo. Una vez adecuadamente fijado, el tejido debe poder resistir las etapas posteriores de procesamiento o de tinción del tejido.

Application

Se ha utilizado disolución de formalina, con taponamiento neutro, al 10% como un fijador histológico general popular para fijar los tejidos.
La disolución de formalina tamponada neutra al 10% se ha utilizado en la tinción rojo aceite O para la diferenciación de la línea celular preadipocito 3T3-L1 y los fibroblastos embrionarios de ratón en adipocitos. Se ha utilizado en análisis histológicos de ojo de conejo e intestino de rata diabética.

    1. The exact buffer concentration for HT511128 is not stated on the website and is considered confidential. The classic textbook formulation for 10% Neutral Buffered Formalin is as follows:

      100 ml Formaldehyde, 37-40%
      900 ml distilled water
      4.0 grams sodium phosphate, monobasic
      6.5 grams sodium phosphate, dibasic (anhydrous)

      The Specification Sheet states the pH range must be between 6.90 to 7.10. The temperature at which the pH is not measured but should be either 20° or 25°C.

    1. Neutral buffered formalin solutions are particularly sensitive to ambient light. Long term exposure to UV light may result in degradation. This product is packaged in natural opaque plastic. Note that solutions should be tightly closed during storage.

    1. It is generally expected that if tissues are left in 10% Neutral Buffered Formalin for extended periods, the tissue may not be suitable for immunofluorescence. However, if the tissue spends only a short time in the 10% NBF, it might still be usable for immunofluorescence.

  5. Is there a 10% neutralized buffered formalin stating the osmolality?

    1. None of the formalin solutions are tested for osmolality. Osmolality is significant for live cells, as cells struggle to survive when their surroundings are vastly different from their own, on the other hand formalin is only a fixative solution. Formaldehyde solutions are buffered to prevent the formation of formalin pigment or acid hematin that forms when tissues are fixed at low pH.

  6. What are the differences between products 1004960700 and HT501128?

    1. Products 1004960700 (Formaldehyde solution, 4% buffered) and HT501128 (10% Neutral Buffered formalin) do not have a substantial difference. With HT501128, the formaldehyde used for dilution has a formaldehyde content of 37 to 40%. When the product is diluted 1 to 10 with water, it is referred to as 10% formalin, which has a formaldehyde content of 4% formaldehyde. Both products have the same concentration of formaldehyde in solution.

  7. Is this product HT501128-4L is RNase/nuclease free?

    1. Currently, none of the formalin solutions are nuclease free. Products that are exo or endo nuclease free are typically labeled as suitable for "molecular" use. If no testing is shown as being performed, the products are not claimed to be nuclease free.
