General description

GAST gene encodes for the protein [Leu15]-Gastrin I in human. In human chromosome, the gene GAST is localized on 17q21.2. G cells in the stomach antrum produces the precursor of [Leu15]-Gastrin I, progastrin. Progastrin undergoes cleavage and processing to yield gastrin, which is trophic for the entire gastrointestinal epithelium. Gastrin is essential for the growth of the digestive system and stimulates the production of gastric acid by parietal cells. Gastrin exerts its function through G-protein-coupled receptor called the cholecystokinin (CCK) or CCK-B receptor (CCK-BR). Gastrin release is stimulated by food, especially protein diet and is inhibited by very low pH. Atrophic gastritis, a Helicobacter pylori infection and long-term administration of proton pump inhibitors infection may cause overexpression of gastrin. Gastric adenocarcinoma show high levels of gastrin.

Application

Gastrin has been used:
  • as a medium supplement establish enteroid cultures
  • as a supplement to standard Dulbecco′s Modified Eagle Medium (DMEM) culture medium, human minigut medium to establish the growth of crypts.

Biochem/physiol Actions

Gastrin I analog with full biological activity and improved stability in aqueous solutions. Leu15 replaces Met15, that is readily oxidized resulting in loss of biological activity.

  1. Could you please provide information regarding the storage duration?

    1. This product is assigned a retest date of 5 years from the date of quality release. A firm expiration date is not assigned. The retest date is lot specific and available in the product Certificate of Analysis. Please see the link below to review a sample or lot specific Certificate:

      The solution stability of this material has not been investigated. In general, peptide solutions may be stable up to 1 week when stored at 2-8°C. Should long term storage be necessary, storing in aliquots at -20°C is suggested. Avoid repeated freeze/thaw cycles. Note that peptides have a tendency to stick to plastic containers upon storage.

  2. Can you reconstitute the powder directly into 0.1 % BSA/PBS?

    1. This product is tested for solubility at 1 mg/ml in 0.1% NaOH or 1% NH4OH. Other solvents have not been tested in-house. This product was lyophilized from 0.1% TFA, which is a strong acid. Reconstituting in 0.1% NaOH would help to neutralize the acid. Reconstituting in PBS will not damage the gastrin, but the solution may still be acidic. There may be instances where the gastrin is more difficult to resuspend in PBS.

  3. Is the powder sterile for cell culture use

    1. This item is not a sterile product. Stock solutions may be filter sterilized.

  4. After preparing aliquots, what is the stability of this product at -20?

    1. The stability of this product in solution has not been evaluated. Generally, peptides should not be stored in solution for extended periods. Should solution storage be necessary, storing aliquots at -20°C is suggested. Note that peptides have a tendency to stick to plastic containers upon storage.

  5. Is this product also suitable for use with other mammalian species, such as mouse, for ex vivo tissue studies, as opposed to just human organoid culture medium?

    1. The product's validation for batch release does not currently cover murine ex vivo tissue testing. However, the following citations may provide insights into its application for ex vivo non-human samples:

  6. I am interested in this product. A colleague uses it for human organoid culture medium. However, I work with murine ex vivo tissues. Is this also applicable for other mammalian species (so mouse in stead of human)?
