General description

Modified Giemsa stain means when the pH of the Giemsa stain is changed from the usual 6.8 to 9.0.[1]

Application

Modified Giemsa stain may be used for the identification of each human chromosome by differentials staining.[1]

Biochem/physiol Actions

When blood films are stained using Giemsa Stain, the nucleus and cytoplasm of white blood cells take on characteristic blue or pink coloration. The use of purified eosin and thiazine dyes minimizes lot-to-lot variation.

Other Notes

Giemsa stain, modified, 0.4%, w/v, in buffered methanol solution, pH 6.8. Also contains stabilizers.

  1. To stain a blood smear does this product need to be diluted in PBS? What is the recommended ratio of stain to PBS?

    1. Giemsa solution should never be diluted in PBS. Giemsa solution is typically diluted with either deionized water or a low molarity phosphate buffer. Weise and Sorsensen phosphate buffer solutions are typically used. Dilutions can range from 1:10 to 1:80. The most common range for dilutions are either 1:20 or 1:40.

    1. Product 32884 has not been suitability tested for G banding of human chromosomes. There is no available protocol for use of the product for G banding of human chromosomes. The quality control testing performed on a lot-to-lot basis includes testing for appearance, density, pH, and UV absorbance.
