Linee guida per il trasferimento di un metodo C18 a Discovery® HS F5:
Di solito sulla HS F5 la ritenzione delle basi dura più a lungo che su una C18. Aumentando dal 5 al 10% il contenuto organico di una separazione C18 di solito si ottiene una ritenzione simile su una HS F5. I risultati con altri composti mostrano un′ampia variabilità. Tuttavia è generalmente vero che i soluti con valori log Po/w inferiori a 2,5 subiscono una ritenzione più prolungata su HS F5 che su una C18. Il grado di differenza dipende fortemente dal soluto.
La fase legata Discovery® HS F5 fornisce separazioni in fase inversa marcatamente differenti rispetto alle colonne C18. Tuttavia, la finestra temporale di ritenzione entro la quale avviene l′eluizione dei composti è la stessa, per cui il trasferimento della maggior parte dei metodi C18 non presenta difficoltà.
