The principal of this kit is based on chemical binding and elution rather than size exclusion. To recover more of the small-sized RNA, an increase the amount of Binding Solution is suggested, however some small RNA molecules, including microRNA, siRNA, tRNA, and 5S rRNA, are not efficiently recovered. To maximize the recovery of small RNAs, refer to Protocol A on page 5 of the user guide – link provided below:

General description
Il kit per RNA totale vegetale Spectrum utilizza una chimica di lisi e di legame e un comodo formato di ′legame-lavaggio-eluizione′ basato su colonna per purificare fino a 100 μg di RNA totale da 100 mg di tessuto in circa 30 minuti. Il rendimento di solito è compreso tra 20 e 60 μg.
Dopo la polverizzazione che riduce il tessuto a una polvere sottile in azoto liquido, le cellule vengono lisate e i detriti cellulari sono separati fisicamente e chimicamente dall′RNA endogeno. L′RNA viene quindi legato a un substrato di silice supportato in colonna e i contaminanti rimasti vengono rimossi attraverso diverse fasi di lavaggio.
L′RNA totale viene eluito dalla colonna e usato nelle applicazioni tipiche, come il Northern blot e la PCR RT e qRT.

We need to know the elution buffer composition for downstream quantification and sequencing. What solution can be used besides the original elution buffer that can have the same proprerties to be used as a blank?
