General description

Application

Phospholipase C (PLC) has been used to study adrenoceptor-mediated transmembrane signaling. It is also used to degrade inositol-containing phospholipids. Furthermore, it has been used to study 9E3 gene activation.

Biochem/physiol Actions

Hydrolyzes the phosphate bond on phosphatidylcholine and other glycerophospholipids yielding diacylglycerol; this enzyme will also hydrolyze the phosphate bonds of sphingomyelin, cardiolipin, choline plasmalogen and ceramide phospholipids.
PLC hydrolyzes the phosphate bond on phosphatidylcholine and other glycerophospholipids yielding diacylglycerol. This enzyme also hydrolyzes the phosphate bonds of sphingomyelin, cardiolipin, choline plasmalogen and ceramide phospholipids. Phospholipase C is induced by thrombin and platelet-activating factor, forming 1,2-diacylglycerol and phosphatidic acid.

Other Notes

One unit will liberate 1.0 μmole of water soluble organic phosphorus from egg yolk L-α-phosphatidylcholine per min at pH 7.3 at 37 °C.

  1. Could you please advise on the recommended buffer for the initial dissolution of P7633? Also, if I prepare a stock solution, is it appropriate to store it directly at -80°C for long-term storage? I would appreciate it if you could provide the recommended

    1. Dissolve this product in cold 50 mM Tris-maleate buffer containing 1% bovine serum albumin at pH 7.3. Prepare the solution immediately before use. Long-term storage of the reconstituted solution at -80 °C has not been validated. For long-term storage, freeze the solution promptly in single-use aliquots, avoid repeated freeze-thaw cycles, and confirm enzyme activity after thawing. Store the unopened lyophilized product at -20 °C.

  2. what is the meaning of 10-50 units/mg protein.

    1. The protein content and enzymatic activity of this product vary from lot to lot. The protein content will range from 20% - 40%, and the activity will range from 10 units - 50 units per milligram of protein. As an example, a batch that reports a protein content of 20% and an activity of 10 units per milligram of protein will have a vial content of 12.5 mg for the 25 unit package size. The protein content and activity are reported in the product Certificate of Analysis. Please see the link below to review a sample or lot specific Certificate:

    1. Unfortunately, the purification method for these products are considered proprietary. However, the following reference may be of interest:

    1. The extraction and purification methods for our Phospholipase C products are proprietary, however higher unit per milligram values indicate a more purified product. Product P7633 contains a specific activity of 10-50 units/mg protein and the protein content is between 20.0 - 40.0 %. Product P4039 will contain a specific activity of ≥150 units/mg protein. The protein content for this product is between 5.0 - 30.0 %. The impact of the methods on the isoform profile of these products is not evaluated. The specific activity of each lot activity will be reported in the Certificate of Analysis. Please view the sample Certificate of Analysis:
