General description

La mucina è composta principalmente da unità di carboidrati, cruciali per l′architettura strutturale e la conformazione. Il monomero della mucina corrisponde a 640 kDa e le proteine contribuiscono soltanto al 20% del peso molecolare. La componente di carboidrati è legata alle catene laterali di treonina e serina. Le mucine sono classificate in mucine secretorie e mucine transmembrana.
Le mucine, o glicoproteine del muco, sono le principali componenti macromolecolari del muco mammifero.[1] La mucina è stata usata anche in uno studio che ha valutato strisci di espettorato concentrato mediante citocentrifugazione per il rilevamento dei bacilli acido-rapidi.[2]

Application

La mucina da stomaco suino è stata utilizzata:
  • nella generazione della curva standard per la quantificazione della mucina-2 generata in condizioni di infiammazione della mucosa nei topi
  • come componente di soluzioni liquide di digestione in modelli di digestione umana in vitro[3]
  • come componente di un fluido simulante la saliva

La mucina da stomaco porcino è stata utilizzata in studi sul sito di legame dell′agglutinina PA-IL da Pseudomonas aeruginosa specifica per il galattosio.[4]

Biochem/physiol Actions

La mucina trova ampio impiego in applicazioni farmacologiche di somministrazione dei farmaci. Le mucine svolgono un ruolo protettivo nell′infiammazione. Lo sregolamento della sintesi delle mucine contribuisce alla tumorigenesi e all′infiammazione cronica.
Il muco forma uno strato protettivo che riveste tutte le superfici epiteliali. I principali componenti strutturali del muco sono le mucine gelificanti. Lo strato di muco che ricopre l′epitelio intestinale contiene come molecola centrale la mucina MUC2. La MUC2 è prodotta dalle cellule caliciformi.[5]

    1. The solubility in water has not been evaluated for these products. It is recommended to follow the reconstitution instructions and reconstitute at a concentration of up to 20 mg/mL in 1 M NaOH.

  2. What is the best way to sterilise the product without destroying the structure?

    1. The structural integrity of this product has not been evaluated post-sterilization. A general procedure for sterilizing mucin has been published as follows:
      1. Place dry powdered mucin in a flask.
      2. Cover the mucin with 95% ethanol.
      3. Heat the covered mucin at 70 °C for 24 hours.
      4. Evaporate the alcohol to recover the mucin.

    1. This product has not been tested in comparison to human-source mucin.

  4. I saw that this product is made by peptide degradation. by that way, can it harvest transmembrane mucins? I wonder this product contains transmembrane mucin too, especially MUC1.

    1. This product is a very crude preparation of mucin obtained by digestion of hog stomach with pepsin, followed by precipitation of the mucin with alcohol. The crude mucin is collected by filtration and dried. It is a mixture of glycoproteins. Transmembrane mucins tend to be heavily glycosylated and are more resistant to digestion however, conditions such as pH, temperature, and activity of the enzyme may lead to degradation. Further characterization of the mucins present in the final product is not performed.

    1. This product is a crude mixture of glycoproteins. The sugar and amino acid composition of the mucin has not been determined.

    1. This crude mucin may not be readily soluble in solution. At 20 mg/mL in 1 M sodium hydroxide, turbid suspensions are observed. To try to increase the solubility, one might try further dilution and sonication, but we have not tried this in our labs.

    1. Product No. M2378 is a crude mucin preparation. Product No. M1778 is a partially purified powder prepared according to the method described in Glenister, D.A. and Salmon. K. Microbial Ecol. in Health & Disease 1, 31, (1988). The amount of bound sialic acids are also different. Please refer to the specification sheets on our website.
