ColorWheel® technology is a novel and proprietary method of creating your own antibody and dye combinations for use in flow cytometry. By incubating any ColorWheel® antibody with any ColorWheel® dye, researchers can quickly and simply produce primary conjugated antibodies for use in single color or multicolor flow cytometry analysis. Each ColorWheel® antibody and dye is lyophilized for long long-term storage, allowing for simplicity and flexibility without compromise. ColorWheel® technology requires both ColorWheel® antibody and ColorWheel® dye for Flow Cytometry application.

Specificità

Clone 3.9 is a mouse monoclonal antibody that detects Integrin alpha-X (CD11c). It targets an epitope within the I-domain from the N-terminal half.

Quality Control Testing:

Evaluated by Flow Cytometry in Human peripheral blood mononuclear cells (PBMC).

Staining of one million human peripheral blood mononuclear cells (PBMC) was performed using 5 μL of a 1:1 mixture of Cat. No. CWA-1056, Anti-Human CD11c (3.9) ColorWheel® Dye-Ready mAb and Cat. No. CWD-PE ColorWheel® Antibody-Ready Phycoerythrin (PE) Dye or the equivalent amount of PE-conjugated Mouse IgG1 isotype control.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user

Integrin alpha-X (UniProt: P20702; also known as CD11 antigen-like family member C; Leu M5; Leukocyte adhesion glycoprotein p150,95 alpha chain; Leukocyte adhesion receptor p150-95; CD11c) is encoded by the ITGAX (also known as CD11C) gene (Gene ID: 3687) in human. CD11c is a single-pass type I membrane glycoprotein that is synthesized with a signal peptide (aa 1-19), which is subsequently cleaved off to generate the mature form that contains a long extracellular domain (aa 20-1107), a transmembrane domain (aa 1108-1128), and a cytoplasmic domain (aa 1129-1163). CD11c is predominantly expressed in macrophages, monocytes, and granulocytes and along with integrin β2 it serves as a receptor for fibrinogen. It recognizes the sequence G-P-R in fibrinogen. The αXβ2 heterodimer binds many ligands, including collagen type 1, iC3b, fibrinogen, ICAM-1, and lipopolysaccharide. It is reported to mediate cell-cell interaction during inflammatory responses and is especially important in monocyte adhesion and chemotaxis. The I domains of CD11 are responsible for the binding of ligands and have a unique structure with 6-7 α-helixes and 6β sheets with interconnecting loops. In CD11c the loops on the I domain are involved in fibrinogen binding and loops α3-α4 and βD-α5 are reported to be more important for recognition of fibrinogen. (Ref.: Vorup-Jensen, T., et al. Stato fisico

Lyophilized from PBS containing D-Mannitol and Sucrose, normal appearance is a dried pellet. Reconstituted antibody solution is stable and functional as assessed by functional testing. Contains no biocide or preservatives, such as azide.

Ricostituzione

1.0 mg/mL after reconstitution at 25 μL in PBS. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Stoccaggio e stabilità

Recommend storage of lyophilized product at 2-8°C. Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial. Reconstitute each vial by adding 25 μL of PBS. Protect from light.
