General description
L'albumina sierica bovina è una proteina usata come modello in vari studi ed è composta da 583 amminoacidi. È una proteina solubile in acqua con un peso molecolare di 66,4 kDa. Sei α-eliche formano i tre domini omologhi della BSA. A seconda del pH, può andare incontro a isomerizzazione conformazionale reversibile. La struttura nativa della proteina diventa reattiva e flessibile in seguito a riscaldamento.
Application
- L′albumina da siero bovino (BSA) è stata usata in soluzioni preparate per l′isolamento di mitocondri cardiaci.
- È stata usata nel tampone delle prove di citotossicità.
- È stata usata per l′induzione di nefropatie nei ratti in seguito a sovraccarico proteico.
Biochem/physiol Actions
Si sospetta che alcuni epitopi conformazionali e di sequenza primaria di BSA siano allergeni nelle allergie umane a carne bovina e latte.
Preparation Note
Chiamato anche frazione di Cohn V, questo prodotto è ottenuto mediante una modifica del processo di Cohn per il frazionamento del plasma a freddo con etanolo.
Ci si riferisce alla sieroalbumina come Frazione V. Questa designazione è presa dal metodo originale introdotto da Cohn per frazionare siero proteine usando la precipitazione in etanolo a freddo. La sieroalbumina è stata trovata da Cohn nella quinta frazione col suo metodo basato sull′etanolo. Da allora il termine “Frazione V” è stato usato da alcuni a prescindere dal metodo di preparazione. Sigma-Aldrich produce e distribuisce sieroalbumine purificate con vari metodi primari inclusi il metodo originale di frazionamento di Cohn, metodi modificati di frazionamento con etanolo, shock termico e cromatografia. Stadi successivi di purificazione possono comprendere cristallizzazione o filtrazione su carbone attivo.

The data sheet lists several methods for further purification of BSA after ethanol precipitation but does not say which of these were used for A4503. Is A4503 charcoal treated? The data sheet is the same for other BSA powders (e.g., A2153).

Is this product biotin-free?

This product is not tested for the presence of biotin. However, Albumin products prepared by cold ethanol fractionation are expected to be biotin-free.

Is Product No. A4053, Albumin from bovine serum, sterile? If not, how would recommend sterilizing it?

This product is not necessarily sterile. Solutions of albumin cannot be autoclaved, so in order to sterilize it we would recommend that it be dissolved in the appropriate medium (water, saline or buffered saline), followed by sterile filtration through a low protein-binding membrane.
