Descrizione generale

Eosin Y, also known as Eosin yellowish is a widely used xanthene dye. It is a counterstain that is used to distinguish between various connective tissue fibers & matrices, and the cytoplasm of different types of cells. Eosin in combination with hematoxylin is useful to demonstrate the general histological architecture of a tissue.

Applicazioni

Eosin Y is suitable for use in hematoxylin and eosin (H&E) staining procedures for various tissue samples like lung nodule and kidney sections.

    1. It is possible that either too much glacial acetic acid was added, or the acetic acid was added too quickly. When using the 0.5% Eosin Y solution sold as catalog number HT110216-500 ml, the instructions recommend adding 0.5 glacial acetic acid slowly to 100 ml of stain solution just prior to use. It's not uncommon to observe this in the lab. In most cases, the precipitate can be resolved by swirling the solution by hand or mixing it on a stir plate.

  3. Is Product 318906 suitable for use in Hematoxylin and Eosin (H&E) staining procedures?

    1. Product 318906 was not intended for use with H&E staining techniques. The dye content for the Eosin Y is 5% in water. For H&E staining, the concentration is typically 0.5 to 1.0 %. At 5 % dye content, the dye is much too concentrated for use with H&E staining. If used at such a high concentration, the cytoplasmic staining is likely to obscure the nuclear staining from the hematoxylin. A well stained H&E slide should show blue nuclear staining and typically at least 3 shades of Eosin staining. When used at 5% dye content, the cytoplasmic staining is likely to be a homogenous or monotone.The 5% Eosin solution has only one common application - determining sperm viability.  In a typical semen analysis, 5% Eosin Y is used in conjunction with 10% Nigrosin. This combination of dyes is used to judge the viability of the sperm. A standard H&E procedure is then used to judge the morphology of the sperm. Sperm counts are typically performed without the use of a dye solution.It is possible to consider a 5% Eosin solution as a 10X staining solution. The product, however, is not suitability tested for use in either a sperm viability procedure or a H&E staining procedure.The following products have been suitability tested for use in H&E staining procedures and are considered suitable for use in In Vitro Diagnostic procedures. They are the preferred products for H&E staining. Most are formulated with 0.5% Eosin Y. The exception is Eosin Y with Phloxine B.
