Description générale

Thioredoxin is a small electron transport protein. The thioredoxin system consists of thioredoxin, thioredoxin-reductase and NADPH. Thioredoxin obtained from E. coli is made up of a single polypeptide chain of 108 amino acids. It has a molecular weight of 11,700. The protein contains no prosthetic group or bound metals.

Spécificité

Specific for natural E. coli and recombinant thioredoxin. It may be used to identify and purify the expression of thioredoxin fusion proteins.

Application

Anti-Thioredoxin−Agarose antibody produced in rabbit has been used for immunoaffinity purification and immunoprecipitation of thioredoxin from bacterial lysates.
Immunofluorescence was carried out on the cerivcal cancer cell lines SiHa, CaSki, and HeLa using an antibody against the redox proteinThioredoxin.

Actions biochimiques/physiologiques

Thioredoxin is a 11.7kDa electron transport protein that functions as a hydrogen donor during the enzymatic reduction of ribonucleotides to yield deoxyribonucleotides. The thioredoxin system is also involved in the enzymatic reductions of methionine sulfoxide and sulfate. The gene encoding E. coli thioredoxin has been cloned into several plasmids and serves as a fusion partner for the expression of other cloned genes. Hence, the system is useful for the production of soluble fusion proteins in the E. coli cytoplasm. Furthermore, in several instances, these fusion proteins fold correctly and thus display full biological activity.

Suspension in 0.01 M phosphate buffered saline, pH 7.4, containing 15 mM sodium azide.

Prepared using antibody coupled to cyanogen bromide activated agarose at 5 mg antibody per mL resin volume.
