Description générale

Tau-tubulin kinase 1 (EC 2.7.11.1; UniProt Q5TCY1; also known as Brain-derived tau kinase) is encoded by the TTBK1 (also known as BDTK, KIAA1855) gene (Gene ID 84630) in human. Tau-tubulin kinase 1 and 2 (TTBK1 and TTBK2) belong to the casein kinase 1 superfamily of kinases. TTBK2 is ubiquitously expressed, while TTBK1 is neuron-specific and can directly phosphorylate tau proteins at multiple Ser/Thr residues that are found on paired helical filament-tau or PHF-tau in Alzheimer′s disease (AD) brains. Consistently, transgenic mice over-expressiong human TTBK1 develop age-dependent pathology in brain, including intraneuronal accumulation of phosphorylated neurofilament (p-NF), tau phosphorylation, cortical and hippocampal astro/microgliosis, calpain I up-regulation, enhanced CDK5/p35 complex formation, down regulation of NMDA receptor, and memory impairment.

Spécificité

Expected to react with both spliced isoforms reported by UniProt (Q5TCY1-1 and Q5TCY1-2). Clone F287-1.1-1E9 detected the full-length as well as the proteolytically processed human and mouse TTBK1 (Sato, S., et al. (Xu, J., et al. (2010). FASEB J. 24(8):2904-2915; 2008). J. Neurosci. 28(53):14511-14521).

Application

Detect TTBK1 using this Anti-TTBK1 Antibody, clone F287-1.1-1E9 validated for use in Western Blotting, Immunohistochemistry.
Immunohistochemistry Analysis: A representative lot detected a much higher TTBK1 immunoreactivity in paraformaldehyde-fixed frozen hippocampus and cortex sections from mice with human TTBK1 transgene (Xu, J., et al. (2010). FASEB J. 24(8):2904-2915).
Immunohistochemistry Analysis: A representative lot detected a much higher TTBK1 immunoreactivity in the subiculum and external pyramidal layer of visual cortex in paraformaldehyde-fixed frozen brain sections from transgenic mice overexpressing human TTBK1 (Sato, S., et al. (2008). J. Neurosci. 28(53):14511-14521).
Western Blotting Analysis: Representative lots detected both the full-length and the proteolytically processed murine TTBK1 in tissues (cerebellum, cortex, spinal cord) from wild-type mice, as well as the expression of human TTBK1 transgene in tissues from transgenic mice (Sato, S., et al. (Xu, J., et al. (2010). FASEB J. 24(8):2904-2915; 2008). J. Neurosci. 28(53):14511-14521).
Western Blotting Analysis: A representative lot detected an upregulation of both the full-length and the proteolytically processed TTBK1 in the frontal cortex gray matter extracts from Alzheimer′s diseased (AD) human brains than age-matched control non-AD brains (Sato, S., et al. (2008). J. Neurosci. 28(53):14511-14521).

Evaluated by Western Blotting in TTBK1 trangenic mouse brain tissue lysate.

Western Blotting Analysis: 2.5 µg/mL of this antibody detected the expression of human TTBK1 transgene in 20 µg of transgenic mouse brain tissue lysate.
