Dimethylsulfoxid (DMSO) ist ein hochpolares organisches Reagenz, das über außergewöhnliche Lösungsmitteleigenschaften für organische und anorganische Chemikalien verfügt.
Dieses Produkt ist als molekularbiologisches Reagenz klassifiziert und ist für molekularbiologische Anwendungen geeignet. Es wurde auf das Vorhandensein von Nukleasen und Proteasen hin analysiert.

Application

DMSO (Dimethylsulfoxid) ist ein hochpolares, aprotisches organisches Lösungsmittel mit vielen Einsatzmöglichkeiten in der organischen Chemie und Molekularbiologie. DMSO wird routinemäßig verwendet in

Polymerase-Kettenreaktion (PCR)
Amplifikation von cDNA-Bibliotheken
DNA-Sequenzierung
Säulen-Ladepuffer für die Poly(A)+ RNA-Selektion
Puffer für die Transformation kompetenter E. coli
Transfektionsprotokolle

Dimethylsulfoxid wird verwendet:

als Lösungsmittel für Oligonukleotide beim Array-Spotting[1]
als Lösungsmittel bei der miRNA-Microarray-Analyse und der quantitativen Echtzeit-PCR (qRT-PCR)
für die Zelllyse während des MTT (3-(4,5-Dimethylthiazol-2-yl)-2,5-Diphenyltetrazolium-Bromid)-Assays
für die Herstellung der Stammlösung während der zytofluorimetrischen Bewertung des MMP (mitochondriales Membranpotential)
bei der Lagerung menschlicher und tierischer Zelllinien und Bakteriophage λ als Kryoprotektor.

Zur Herstellung einer steril gefilterten DMSO-Lösung wird die Verwendung einer PTFE- oder Nylonmembran empfohlen. Membranen aus Celluloseacetat werden nicht empfohlen.

May I know the concentration (mg/ml) for DMSO (cat no. D8418)?

  This product is supplied as a neat or pure liquid with a density of 1.10 g/mL and a molecular weight of 78.13 g/mol. The minimum purity is 99.9%, thus the molarity will vary slightly from lot to lot. The molarity is 14.1 M.

  Yes, Dimethyl sulfoxide (DMSO) is generally considered to be compatible with Polyethylene terephthalate (PET) material. The following information can be found on the Product Data Sheet:
  -Compatible: LDPE, HDPE, polypropylene, PPCO (polypropylene copolymer), polymethylpentene, nylon, teflon FEP
  -Moderately compatible: polystyrene, ECTFE/ETFE
  -Incompatible: polysulfone, flexible and rigid PVC tubing, polycarbonate

Is this product sterile?

  This product is not sterile. To prepare a sterile filtered DMSO solution, it is recommended to use a Teflon, polypropylene, or nylon membrane. See products D2650 or D2438 for sterile DMSO product options. Please review these products at the links provided below:

What can I use to solubilize Product D8418, Dimethyl sulfoxide?

  This product is miscible in water (1 mL DMSO + 1 mL H2O), yielding a clear, colorless solution. DMSO is a very hygroscopic liquid and should protected from exposure to moisture. DMSO is also miscible with ethanol, acetone, ether, benzene, and chloroform.

How does the storage temperature relate to shipping conditions?

  The storage conditions that a Sigma-Aldrich catalog and label recommend for products are deliberately conservative. For many products, long-term storage at low temperatures will increase the time during which they are expected to remain in specification and therefore are labeled accordingly. Where short-term storage, shipping time frame, or exposure to conditions other than those recommended for long-term storage will not affect product quality, Sigma-Aldrich will ship at ambient temperature. The products sensitive to short-term exposure to conditions other than their recommended long-term storage are shipped on wet or dry ice. Ambient temperature shipping helps to control shipping costs for our customers. At any time, our customers can request wet- or dry-ice shipment, but the special handling is at customer expense if our product history indicates that the product is stable for regular shipment.

  DMSO has been used to oxidize thiols and disulfides to sulfonic acids, hydrolysis of epoxides, thioalkylation of phenols, and oxidation of primary alcohols and halides, and esters of primary alcohols to aldehydes.
