Itraconazole EP Reference standard, intended for use in laboratory tests only as specifically prescribed in the European Pharmacopoeia.

Biochem/physiol Actions

Itraconazol hemmt Cytochrom-P-450-abhängige Enzyme, was zur Inhibition der Ergosterol-Synthese führt. Das geschieht durch Interaktion mit 14-α-Demethylase, einem Cytochrom-P-450-Enzym, das für die Umwandlung von Lanosterol in Ergosterol benötigt wird. Ergosterol ist ein entscheidender Bestandteil pilzlicher Zellmembranen. Deshalb führt seine Hemmung zu erhöhter Zelldurchlässigkeit, sodass Zellinhalte austreten können. Itraconazol kann auch die endogene Atmung hemmen, mit Membran-Phospholipiden interagieren, die Umwandlung von Hefen in Mycelformen hemmen, die Purin-Aufnahme hemmen und die Biosynthese von Triglyceriden und Phospholipiden beeinträchtigen.

Synthetisches Breitspektrum-Triazol-Antimykotikum. Wirkungsweise: Hemmt Cytochrom-P450-abhängige Enzyme einschließlich 14α-Demethylase. Die Inhibition führt dazu, dass die Biosynthese von Ergosterol verhindert wird, einem kritischen Bestandteil der Zellwand von Pilzen.
