General description

L'aprotinine de poumon de bovin est un polypeptide monomère globulaire d'un poids moléculaire de 6,5 kDa. Couramment utilisée comme inhibiteur de protéases à sérine non spécifique, l'aprotinine comporte un feuillet β antiparallèle, une hélice 310 N-terminale et une hélice α C-terminale. Les résidus d'acides aminés 13 à 18 de l'aprotinine sont indispensables à sa fixation sur les protéases à sérine.

Application

L'aprotinine est largement utilisée comme inhibiteur de la trypsine.
L'aprotinine de poumon de bovin a été utilisée :
  • Comme inhibiteur de protéases dans un tampon d'analyse par radioimmunoprécipitation (RIPA) pour l'homogénéisation de cellules endothéliales microvasculaires cardiaques (CMEC)(4) et de cellules épithéliales mammaires
  • Dans une étude sur l'angiogénèse de fibroblastes
  • Pour la stabilisation protéomique d'un surnageant salivaire

Biochem/physiol Actions

L'aprotinine inhibe les protéases telles que la trypsine, la plasmine, la chymotrypsine et la thrombine. Elle bloque la synthèse de la bradykinine à partir du kininogène. Elle est utile dans le traitement des pertes sanguines en cours d'intervention chirurgicale.
L'aprotinine est un inhibiteur compétitif de sérine protéase qui forme des complexes stables avec les sites actifs de l'enzyme et les bloque. Cette fixation est réversible, et la plupart des complexes aprotinine-protéase se dissocient aux pH extrêmes (> 10 ou < 3). En terme de structure, l'aprotinine est une protéine globulaire monomère issue de poumon de bovin, constituée de 58 acides aminés disposés de façon à former une chaîne polypeptidique unique comportant trois réticulations (ponts disulfure).

Preparation Note

Ce produit est une poudre lyophilisée présentant une activité de 3-8 TIU/mg de poudre solide. L'aprotinine est facilement soluble dans l'eau (> 10 mg/ml) et dans les tampons aqueux de faible force ionique. Les solutions diluées ont tendance à être moins stables que les solutions concentrées, même si la stabilité en solution dépend également du pH. L'aprotinine est relativement stable vis-à-vis de la dénaturation ; seule la thermolysine s'est avérée capable de la dégrader à 60-80 °C. La stérilisation de l'aprotinine peut être effectuée par filtration à l'aide d'un filtre de 0,2 μm.

Other Notes

Une unité d'inhibiteur de trypsine (TIU) réduit de 50 % l'activité de deux unités de trypsine, une unité de trypsine hydrolysant 1,0 μmole de p-nitroanilide de N-alpha-benzoyl-DL-arginine par minute à pH 7,8 et 25 °C. Une autre unité fréquemment rencontrée est la KIU : 1 TIU = 1 300 KIU.
