Clone 1J6 is a ZooMAb® Rabbit recombinant monoclonal antibody that specifically detects Solute carrier family 12 member 2 (SLC12A2). It targets an epitope within 19 amino acids from the C-terminal, cytoplasmic domain.

Application

Quality Control Testing

Evaluated by Western Blotting in HeLa cell lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected SLC12A2 in HeLa cell lysate.

Tested Applications

Western Blotting Analysis: A 1:10,000 dilution from a representative lot detected SLC12A2 in Human and Mouse brain tissue lysates.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected SLC12A2 in human pancreas tissue sections.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected SLC12A2 in HeLa cells.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Description de la cible

Solute carrier family 12 member 2 (UniProt: P55011; also known as Basolateral Na-K-Cl symporter, Bumetanide-sensitive sodium-(potassium)-chloride cotransporter 2, SLC12A2) is encoded by the SLC12A2 (also known as NKCC1) gene (Gene ID: 6558) in human. SLC12A2 belongs to the subfamily of cation-chloride cotransporters that mediate electroneutral transport of sodium, potassium, and chloride across the plasma membrane. It is a multi-pass membrane protein with seven cytoplasmic domains, sixteen transmembrane domains (including two discontinuous), and six extracellular domains. It is expressed in many tissues and plays a vital role in the regulation of ionic balance and cell volume. Its activity is inhibited by bumetanide and furosemide-type of compounds. It has a major impact on intracellular chloride ion concentration in vascular smooth muscle cells (VSMC) and in neuronal cells. In primary hypertension, its activity is increased in VSMC and paraventricular nucleus neurons that leads to elevated peripheral resistance in systemic circulation and activation of sympathetic nervous system. Mutations in SLC12A2 gene have been linked to antenatal Bartter syndrome 1, an autosomal recessive disorder that is characterized by impaired salt reabsorption in the thick ascending loop of Henle, salt wasting, hypokalemic metabolic alkalosis, and hypercalcinuria. Some mutations are also associated with neurodevelopment disorders and hearing loss. Two isoforms of SLC12A2 have been described that are produced by alternative splicing. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Forme physique

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Reconstitution

300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Stockage et stabilité

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
