Immunohistomount (I1161) is suitable for mounting IHC slides stained using DAB as the chromogen. Unlike Fast Red or AEC, which are alcohol-soluble, DAB is not soluble in either alcohol or in clearing agents such as xylene or xylene substitutes. Most other permanent mounting medias are also suitable for use, such as DPX, NeoMount, Entellen, Eukit, and Permount.

There are two ways to look at slides under an oil immersion lens: 1. For oil immersion lens, a drop of oil can be placed directly on dried ImmunoHistoMount mounting medium for visualization. After use, the oil can be removed by wiping with Kimwipes. 2. Apply 2-3 drops of any organic mounting medium on top of dried ImmunoHistoMount mounting medium; which has protected the specimen, coverslip slide with coverslip; the slide is ready for visualization.

How does the storage temperature relate to shipping conditions?

The storage conditions that a Sigma-Aldrich catalog and label recommend for products are deliberately conservative. For many products, long-term storage at low temperatures will increase the time during which they are expected to remain in specification and therefore are labeled accordingly. Where short-term storage, shipping time frame, or exposure to conditions other than those recommended for long-term storage will not affect product quality, Sigma-Aldrich will ship at ambient temperature. The products sensitive to short-term exposure to conditions other than their recommended long-term storage are shipped on wet or dry ice. Ambient temperature shipping helps to control shipping costs for our customers. At any time, our customers can request wet- or dry-ice shipment, but the special handling is at customer expense if our product history indicates that the product is stable for regular shipment.

What is the step-by-step procedure for using ImmunHistoMount (Product No. I1161)?
1 answer-
1. Bring the vial to room temperature. 2. Rinse slide to be mounted with Distilled or Deionized water, then touch the edges of slide on a paper towel to remove excess water. Place slides on a flat surface. 3. Turn the vial upside down and open the dropper to remove any air bubbles. 4. Apply 3-4 drops of mounting medium directly on top of the specimen and spread out evenly by tilting the slide back and forth to spread evenly, or use a 0.2 ml plastic pipette tip to evenly distribute the mounting media, making sure the tissue is not touched. Excess medium can be removed by touching the edges of the slide against a paper towel. 5. Let stand at room temperature for about 1 to 2 hours. Alternatively, the slides can be heated at 37-40°C for 40-60 minutes. After setting, the slides are ready for visualization under a microscope.
