Application

La 5-bromo-2′-désoxyuridine (5-BrdU) est un analogue de la thymidine incorporé dans l'ADN. La 5-BrdU est couramment et abondamment utilisée pour mesurer la synthèse de l'ADN et marquer les cellules en division. Ainsi, la 5-BrdU permet d'étudier la signalisation cellulaire ainsi que d'autres processus qui induisent la prolifération cellulaire.
La 5-bromo-2′-désoxyuridine a été utilisée :
  • comme supplément pour étudier l'effet de crises électroconvulsives sur la neurogenèse de l'hippocampe
  • comme supplément pour étudier la fonction de perte de gènes dans le cœur du poisson-zèbre adulte
  • dans un test de la prolifération des lymphocytes T{133}

Biochem/physiol Actions

Il s'agit d'un analogue de la thymidine utilisé comme agent mutagène en recherche génétique. Il permet une incorporation sélective dans l'ADN cellulaire au cours de la phase S.
La 5-bromo-2′-désoxyuridine (BrdU) est utilisée dans l'analyse de la prolifération cellulaire, car elle est facile à incorporer dans l'ADN au cours de la phase S du cycle cellulaire. La BrdU stimule la différenciation et la maturation cellulaires dans les lignées de cellules leucémiques tandis qu'elle inhibe la différenciation des cellules érythroleucémiques de Friend. Les études sur l'incorporation de la BrdU utilisent souvent des anticorps spécifiques de la BrdU marqués par fluorescence pour détecter la BrdU incorporée par des techniques telles que la cytométrie en flux ou la microscopie de fluorescence.

  1. Could specific insights into the use of this product be provided? BrdU was administered in water to mice for up to 2 weeks for an in vivo cell proliferation assay. However, upon staining with an anti-BrdU antibody, no BrdU incorporation was detected in the positive control (small intestine), despite trying various concentrations. Any information on the particular use of this product would be much appreciated

    1. Given the BrdU incorporation into DNA of cells, using the Western blot method for detection would be quite challenging. Typically, the immunohistochemistry method is employed for this purpose. When it comes to the application on small intestine samples, the following citation of B5002 via immunohistochemical detection may be useful for protocol reference:

  2. How long can BrdU be stored at -20 degrees and -4 degrees, after being diluted in sterile saline and being filtered with a 0.22 µm filter?

    1. BrdU should be stored at –20 oC and desiccated. Stock solutions can be made in DMSO to up to 50 mg/mL, as indicated in the Product information sheet. The stock can be stored for 6 months at -20°C in single use aliquots.

      No information is available on storage conditions for BrdU dissolved in saline.
