Clone 1J18 is a ZooMAb® rabbit recombinant monoclonal antibody that specifically detects Homeobox protein cut-like 1 (CUX1). It targets an epitope within 21 amino acids from the C-terminal half.

KLH-conjugated linear peptide corresponding to 21 amino acids from the C-terminal half of human Homeobox protein cut-like 1 (CUX1).

Application

Quality Control Testing

Evaluated by Western Blotting in HEK293 cell nuclear extract.

Western Blotting Analysis: A 1:10,000 dilution of this antibody detected CUX1 in HEK293 cell nuclear extract.

Tested Applications

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected CUX1 in Human cerebellum tissue sections.

Affinity Binding Assay: A representative lot of this antibody bound CUX1 peptide with a KD of 6.7 x 10-8 in an affinity binding assay.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected CUX1 in K562 cells.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Homeobox protein cut-like 1 (UniProt: P39880; also known as CCAAT displacement protein, CDP, Homeobox protein cux-1, CCAAT displacement protein, CDP, Homeobox protein cux-1) is encoded by the CUX1 (also known as CUTL1) gene (Gene ID: 1523) in human. Homebox protein CUX1 is member of the CUT Homebox family that has a broad role in mammalian development. It is a transcription factor that is involved in the control of neuronal differentiation in the brain. It regulates dendrite development and branching, and dendritic spine formation in cortical layers II-III. It has a conserved coiled-coil region at the N terminus (aa 56-407), three internal 60-amino-acid repeats (Cut repeats; 542-629; 934-1021; and 1117-1204), and a divergent homeodomain located near the C terminus (aa 1244-1303). CUX1 serves as a repressor of developmentally regulated gene expression and acts by preventing binding of positively-acting CCAAT factors to promoters. It is also a component of NF-muNR repressor, which binds to the matrix attachment regions (MARs) (5′ and 3′) of the immunoglobulin heavy chain enhancer. CUX1 is shown to repress T-cell receptor β enhancer function by binding to the matrix attachment region beta (MARβ), which is an ATC-rich DNA sequence located upstream of the TCR β enhancer. Its cleaved product, CDP/Cux p110, is reported to play a role in cell cycle progression, in particular at the G1/S transition. While CUX1 only transiently binds to DNA and carries the CCAAT-displacement activity, CDP/Cux p110 makes a stable interaction with DNA and stimulates expression of genes such as POLA1. Eleven different isoforms of CUX1 have been described that are produced by alternative splicing. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, Affinity, reproducibility, and stability over conventional monoclonals. Forme physique

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Reconstitution

300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Stockage et stabilité

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
